Article Synopsis
  • Rhomboid proteases, including human RHBDL4, are important due to their roles in protein degradation and disease, particularly cancer, but effective chemical tools to study them have been lacking.
  • An activity-based protein profiling (ABPP) assay was developed specifically for RHBDL4, revealing that it undergoes processing to form active proteoforms with shorter C-termini in cells.
  • The study also identified specific chemical compounds that can inhibit RHBDL4 and showed that there are differences in inhibitor preferences between RHBDL4 and another rhomboid protease called PARL, highlighting the utility of ABPP for enzyme characterization and selective targeting.

Triple-negative breast cancer (TNBC) is associated with metabolic heterogeneity and poor prognosis with limited treatment options. New treatment paradigms for TNBC remains an unmet need. Thus, therapeutics that target metabolism are particularly attractive approaches.

Exposure to heavy metals, including cadmium (Cd), can induce neurotoxicity and cell death. Cd is abundant in the environment and accumulates in the striatum, the primary brain region selectively affected by Huntington's disease (HD). We have previously reported that mutant huntingtin protein (mHTT) combined with chronic Cd exposure induces oxidative stress and promotes metal dyshomeostasis, resulting in cell death in a striatal cell model of HD.

Resuming elective surgery amid shortages of rapid severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) tests and personal protective equipment, while protecting patient and staff safety, posed a challenge at the study institution. Many hospital systems implemented testing of all presurgical patients, using results from tests performed 3 or more days prior to surgery. Among asymptomatic persons, the percent positive rates of coronavirus disease 2019 (COVID-19) tests in the region did not appear to justify this practice.
