Progress for ocular AAV gene therapy has been hindered by AAV-induced inflammation, limiting dose escalation and long-term efficacy. Broadly, the extent of inflammatory responses alters with age and sex, yet these factors are poorly represented in pre-clinical development of ocular AAV gene therapies. Here, we combined clinical imaging, flow cytometry and bulk-sequencing of sorted microglia to interrogate the longitudinal inflammatory response following intravitreal delivery of AAV2 in young (3-month), middle aged (9-month) and old (18-month) Cx3cr1-creER:R26tdTomato+/- mice of both sexes.

Novel π-expanded and N-doped fluoranthenes were synthesized and thoroughly investigated. These eight unsubstituted compounds are obtained in a multistep synthesis with CH activation as the last key reaction step. The structures vary in their position of π-expansion on the fluoranthene scaffold and the location of the pyridinic nitrogen atom.

Purpose: To analyze the national epidemiologic trends of squamous cell carcinoma of the conjunctiva (SCCC) over 4 decades, providing the largest report on disease-specific survival (DSS) outcomes and investigating the risk of developing second primary neoplasms in these patients.

Methods: This retrospective, population-based cohort study analyzed 1,172 patients diagnosed with SCCC from January 1, 1975, through December 31, 2016. Data were extracted from the US Surveillance, Epidemiology, and End Results database.

Article Synopsis
  • A new method for creating enantiomerically enriched (S)-α-aminopropionic acid derivatives was developed using a reaction called Cu(I) catalyzed [3 + 2] cycloaddition, which combines azides with alkynes.
  • The process started with a specific nickel complex and involved several key chemical components, including Schiff bases and phenyl azides.
  • The absolute configuration and purity of the resulting amino acids were confirmed through advanced techniques like circular dichroism and chiral HPLC analysis, leading to the successful synthesis of seven new non-proteinogenic (S)-α-amino acids.

Article Synopsis
  • The study aimed to analyze trends in ocular trauma from motor vehicle accidents (MVAs) involving the drivers between 2007 and 2014 using national trauma data.
  • Results showed a 25.3% increase in MVA-associated ocular injuries over the eight years, with men representing the majority of cases and orbital floor fractures being the most common type of injury.
  • Although the overall mortality rate was 4.3%, older adults faced higher risks, especially regarding open globe injuries and other severe ocular trauma.
