We have applied an automated real-time quantitative PCR assay using a double-labeled fluorogenic probe to detect t(9;22)-positive cells in haematological malignancies. The results are expressed as the ratio of chimeric bcr-abl transcripts on abl transcripts. Highly reproducible results were obtained for t(9;22)-positive K562 RNA.

We present an approach for designing a knowledge-based system, called Sequence Acquisition In Context (SAIC), that will be able to cooperate with a biologist in the analysis of DNA sequences. The main task of the system is the acquisition of the expert knowledge that the biologist uses for solving ambiguities from gel autoradiograms, with the aim of re-using it later for solving similar ambiguities. The various types of expert knowledge constitute what we call the contextual knowledge of the sequence analysis.

Based on consideration of rat plasma Ca and 45Ca concentrations, we analyze the circadian behavior of Ca metabolism of the rat as the temporal expression of a self-organized system. We present a self-oscillatory model M for rat Ca metabolism based on a compartmental formalism, which includes a second-order autocatalytic process. M describes the entire mass of Ca as made up of eight compartments and predicts a distinction between 1) the amount of Ca deposited in zones of rapid bone growth and reutilized during bone maturation and 2) the amount of Ca in mature bone subdivided into four compartments.

The experimental data obtained from in vivo single pass perfusion of duodenal, jejunal, and ileal intestinal segments of 33- and 50-day-old rats have been used to test a series of models for calcium absorption. Each model was checked for the statistical validity and goodness-of-fit with the experimental data. The model adopted for the duodenum and jejunum had two major components, one saturable and the other nonsaturable, and a minor secretory component.
