The Antarctic Circumpolar Current (ACC) dominates the transfer of heat, salt, and tracers around the Southern Ocean (SO), driving the upwelling of carbon-rich deep waters around Antarctica. Paleoclimate reconstructions reveal marked variability in SO circulation; however, few records exist coupling quantitative reconstructions of ACC flow with tracers of SO upwelling spanning multiple Pleistocene glacial cycles. Here, we reconstruct near-bottom flow speed variability in the SO south of Africa, revealing systematic glacial-interglacial variations in the strength and/or proximity of ACC jets.

Article Synopsis
  • Bulk deconvolution using single-cell RNA-seq is important for analyzing complex biological samples, but differences in sequencing technology can lead to inaccuracies.
  • The researchers created a new framework called DeMixSC that improves deconvolution by utilizing well-matched benchmark datasets to account for these technological discrepancies.
  • DeMixSC was tested on datasets from healthy retinas and ovarian cancer tissues, showing improved accuracy and the ability to reveal significant biological differences in patient groups, highlighting its usefulness in clinical applications.

Multiomic profiling of single cells by sequencing is a powerful technique for investigating cellular diversity. Existing droplet-based microfluidic methods produce many cell-free droplets, underutilizing bead barcodes and reagents. Combinatorial indexing on microplates is more efficient for barcoding but labor-intensive.

Background: The role of computed tomography (CT) before lumbar puncture (LP) is unclear, with limited evidence for a causal link between LP and cerebral herniation or for the ability of CT to identify individuals at risk of herniation. The risks of LP delay or deferral are potentially greater in high-HIV prevalence, resource-limited settings; we analyzed data from such a setting to determine the impact of CT on time to LP and treatment, as well as mortality.

Methods: Adults with suspected central nervous system (CNS) infection were enrolled prospectively into the Botswana National Meningitis Survey between 2016 and 2019.

Background: Gay and bisexual men (GBM) remain overrepresented among syphilis diagnoses in Australia and globally. The extent to which changes in sexual networks associated with HIV pre-exposure prophylaxis (PrEP) and treatment as prevention (TasP) may have influenced syphilis transmission among GBM at the population-level is poorly understood. We describe trends in syphilis testing and incidence among GBM in Australia over eleven years spanning widespread uptake of HIV PrEP and TasP.

Article Synopsis
  • Australia implemented a national HPV vaccination program for girls in 2007 and boys in 2013, achieving strong coverage among both groups, with a study focusing on HPV prevalence among men aged 18-35 based on their vaccination status.
  • Of the 1,625 men studied, the HPV prevalence was similar for vaccinated and unvaccinated men who have sex with women (10.6% vs. 10.7%), while unvaccinated men who have sex with men had a significantly higher prevalence (40.3%) compared to vaccinated men (29.9%).
  • The findings highlight that universal vaccination is crucial for all adolescents, particularly to ensure that men who have sex with men also gain protection from HPV, as older

To efficiently yet reliably represent and process information, our brains need to produce information-rich signals that differentiate between moments or cognitive states, while also being robust to noise or corruption. For many, though not all, natural systems, these two properties are often inversely related: More information-rich signals are less robust, and vice versa. Here, we examined how these properties change with ongoing cognitive demands.

Chronic obstructive pulmonary disease (COPD) is a common lung disease that negatively affects health-related quality of life (QoL). Utility values, which measure QoL by weighting health states with societal preferences, are required for the cost-utility models that drive economic evaluations and policy decisions. Moayeri et al.

Objective: Intergenerational studies have identified relations between adolescents' and their future offspring's cannabis and alcohol use, but rarely have examined the association for other illicit drug use. Given the low prevalence of such use in community populations, we pooled data from three prospective intergenerational studies to test this link.

Method: Participants were 1,060 children of 937 parents who had been repeatedly assessed since early adolescence.

Objective: Neonatal endotracheal intubation is a lifesaving but technically difficult procedure, particularly for inexperienced operators. This secondary analysis in a subgroup of inexperienced operators of the Stabilization with nasal High flow during Intubation of NEonates randomised trial aimed to identify the factors associated with successful intubation on the first attempt without physiological stability of the infant.

Methods: In this secondary analysis, demographic factors were compared between infants intubated by inexperienced operators and those intubated by experienced operators.

The COVID-19 lockdowns saw many individuals lose income, experience distress and increase intake of foods that would typically be considered less 'healthy' (more processed and less fresh produce). Establishing whether there are direct and indirect links between these variables would be of benefit in preparing for similar future events but also has implications for the current global financial climate, where many are experiencing relative decreases in income. Adults in two locations (UK and Australia) (N = 917) completed online questionnaires to explore the impact of the first COVID-19 lockdown on their change in income, emotional wellbeing (depression, anxiety, stress, loneliness), resilience and diet quality.

Nonsense and missense mutations in the transcription factor PAX6 cause a wide range of eye development defects, including aniridia, microphthalmia and coloboma. To understand how changes of PAX6:DNA binding cause these phenotypes, we combined saturation mutagenesis of the paired domain of PAX6 with a yeast one-hybrid (Y1H) assay in which expression of a PAX6-GAL4 fusion gene drives antibiotic resistance. We quantified binding of more than 2700 single amino-acid variants to two DNA sequence elements.

The survival and health of preterm and critically ill infants have markedly improved over the past 50  years, supported by well-conducted neonatal research. However, newborn research is difficult to undertake for many reasons, and obtaining informed consent for research in this population presents several unique ethical and logistical challenges. In this article, we explore methods to facilitate the consent process, including the role of checklists to support meaningful informed consent for neonatal clinical trials.

Chronic pain is one of the most significant health issues in the United States, affecting more than 20% of the population. Despite its contribution to the increasing health crisis, reliable predictors of disease development, progression, or treatment outcomes are lacking. Self-report remains the most effective way to assess pain, but measures are often acquired in sparse settings over short time windows, limiting their predictive ability.
