The achievement of complete remission (CR) is crucial for acute myeloid leukemia (AML) patients undertaking curative therapy, but relapse often occurs within months, highlighting the need for strategies to prolong disease-free survival (DFS). Our phase III study compared the efficacy and safety of azacitidine (AZA) to best supportive care (BSC) in elderly AML patients who achieved CR following intensive induction and consolidation therapy. This ancillary study (QOL-ONE Trans-2) evaluated biological changes in bone marrow using Next-Generation Sequencing (NGS).

SOX17 has recently emerged as a novel immunohistochemical marker for cancers of endometrial and ovarian origin with improved specificity compared with the widely used Mullerian marker PAX8. However, evaluation of SOX17 in benign and malignant peritoneal mesothelial proliferations remains limited, and these may mimic gynecologic carcinomas, particularly on small biopsies. We evaluated SOX17 and PAX8 expression in 20 benign mesothelial lesions (5 adenomatoid tumors, 5 well-differentiated papillary mesothelial tumors, and 10 peritoneal inclusion cysts) and 16 epithelioid peritoneal mesotheliomas.

A bimetallic plasmonic nanoparticles-based approach for the untargeted evaluation of phenolic compounds (PC)-pattern and antioxidant capacity (AoC) is proposed. The rationale relies on the PC's ability to drive the formation of bimetallic silver/gold nanocolloidal 'probes' with different conformations. Ag/Au bimetallic nanostructures, according to the PCs' amount and class, return characteristic plasmonic and colorimetric tags.

Lupin seeds are recognized for their nutritional value and potential health benefits, but they contain also a significant amount of alkaloids, an anti-nutritive class of compounds, which vary significantly in composition within and between species due to environmental factors. In this study, we developed a predictive multi-experiment approach using ultra-high performance liquid chromatography coupled with triple quadrupole with linear ionic trap tandem mass spectrometry (UHPLC-QqQ-LIT-MS/MS) for comprehensive alkaloid profiling and geographical classification of L. samples originating from four different Italian regions.

STK11 adnexal tumor is a novel malignant neoplasm of uncertain histogenesis frequently arising in a para-adnexal location and associated with Peutz-Jeghers syndrome in ∼50% of patients. Its broad morphologic spectrum and nonspecific immunohistochemical profile has resulted in misclassification in the past as a variety of other neoplasms including those of wolffian, sex cord-stromal, mesothelial, and epithelial derivation. This review focuses on the spectrum of adnexal neoplasms that may develop in Peutz-Jeghers syndrome, with particular emphasis on STK11 adnexal tumor and its differential diagnosis.

  • Spindle cell-rich testicular sex cord-stromal tumors (TSCSTs) include types like myoid gonadal stromal tumor (MGST), adult granulosa cell tumor (AGCT), and unclassified TSCST, showing histopathologic similarities and lacking clear oncogenic drivers.
  • DNA sequencing indicates common chromosomal gains in these tumors, but further validation of these findings has not yet taken place, and the amount of chromosomal alterations varies among individual tumors.
  • This study used fluorescence in-situ hybridization (FISH) to assess chromosomal changes in 10 tumors, revealing consistent increases in specific chromosomes across various cases, while also uncovering partial disagreements between FISH analysis and previous genomic sequencing results,

Endocervical adenocarcinomas (EACs) are a group of malignant neoplasms associated with diverse pathogenesis, morphology, and clinical behavior. As a component of the International Society of Gynecological Pathologists International Endocervical Adenocarcinoma Project, a large international retrospective cohort of EACs was generated in an effort to study potential clinicopathological features with prognostic significance that may guide treatment in these patients. In this study, we endeavored to develop a robust human papillomavirus (HPV)-associated EAC prognostic model for surgically treated International Federation of Gynecology and Obstetrics (FIGO) stage IA2 to IB3 adenocarcinomas incorporating patient age, lymphovascular space invasion (LVSI) status, FIGO stage, and pattern of invasion according to the Silva system (traditionally a 3-tier system).

High power femtosecond laser pulses launched in air undergo nonlinear filamentary propagation, featuring a bright and thin plasma channel in air with its length much longer than the Rayleigh length of the laser beam. During this nonlinear propagation process, the laser pulses experience rich and complex spatial and temporal transformations. With its applications ranging from supercontinuum generation, laser pulse compression, remote sensing to triggering of lightning, the underlying physical mechanism of filamentation has been intensively studied.

Peritoneal inclusion cysts (PICs) are unilocular or multilocular cystic lesions lined by bland mesothelial cells. While most are small and localized, rare examples may be large or multifocal with diffuse peritoneal involvement, causing clinical and even pathologic concern for malignancy. We examined 20 PIC, including 8 large solitary and 12 multifocal lesions.

Uterine inflammatory myofibroblastic tumors (IMTs) are rare mesenchymal neoplasms of uncertain malignant potential. Aside from the recently described risk stratification score, which has not been validated by other studies, and rare reports of aberrant p16 expression in malignant tumors, there are no criteria to reliably predict behavior. Herein, we evaluated the clinicopathologic features and p16 expression patterns in 30 IMTs, with genomic profiling performed in a subset (13 malignant, 3 benign).

Article Synopsis
  • A study was conducted on the genomic profiles of uterine smooth muscle tumors, focusing on two types: fumarate hydratase deficient leiomyomas and leiomyomas with bizarre nuclei, to understand their relationship with clinical outcomes.
  • All patients in the study were alive without disease after an average follow-up of 87.3 months, leading to the conclusion that certain genomic indices (GI) were not reliable indicators of malignancy in these tumor types.
  • The research identified three groups of tumors based on genomic alterations and their associated GIs, suggesting that a GI less than 10 indicates benignity, while a GI greater than 10 does not necessarily indicate malignancy, offering a new diagnostic approach for these tumors

We review the time honored but still frequently challenging features of ovarian sex cord-stromal tumors and also emphasize new developments, including unusual morphologic appearances that, despite the relative rarity of many of the tumors, result in a disproportionate number of differential diagnostic problems, variant immunohistochemical profiles, and specific molecular and syndromic associations. These neoplasms are also of historical interest as current knowledge is still based in significant part to the contributions of 2 giants of gynecologic pathology, Dr Robert Meyer and Dr. Robert E.

In the era of personalized medicine there is an increasing need for the assessment of patient-reported outcomes (PROs) to become a standard of patient care. Patient-reported outcome measures (PROM) are important in assessing significant and meaningful changes as a result of an intervention based on a patient's own perspective. It is well established that active multiple myeloma (MM) can be characterized by a high burden of disease and treatment-related symptoms, with considerable worsening of quality of life (QoL).

Aims: NTRK-rearranged sarcomas of the female genital tract mainly occur in the uterus (more commonly cervix than corpus) and are characterized by a "fibrosarcoma-like" morphology and NTRK gene rearrangements. These neoplasms may exhibit histological overlap with other entities and can present diagnostic difficulties without molecular confirmation. Pan-TRK immunohistochemistry was developed to identify tumours harbouring NTRK rearrangements.
