Article Synopsis
  • Diphtheria toxin (DT) is crucial for diagnosing diphtheria, making its detection in corynebacterial cultures essential for lab tests.
  • A new immunoassay using monoclonal antibodies (mAbs) has been developed to measure the level of DT production, enabling highly sensitive detection via sandwich ELISA.
  • The ELISA method has shown high diagnostic sensitivity (99%) and specificity (100%), and it reveals that certain strains produce significantly less DT than others, paving the way for more efficient diphtheria diagnosis techniques.

xMAP technology was used for simultaneous identification of six protein toxins (staphylococcal enterotoxins A and B, cholera toxin, ricin, botulinum toxin A, and heat labile toxin of E. coli). Monoclonal antibody-conjugated xMAP microspheres and biotinilated monoclonal antibodies were used to detect the toxins in a sandwich immunoassay format.
