Background: S-1 monotherapy had previously been widely used as a second-line treatment for pancreatic cancer (PC) after gemcitabine-based chemotherapy mainly in Japan. Based on the results of the NAPOLI-1 trial, the recommended second-line therapy is now liposomal irinotecan plus fluorouracil/folinic acid (nal-IRI + 5-FU/LV). However, there have been no studies comparing nal-IRI + 5-FU/LV therapy with S-1 monotherapy.

Tracheal reconstruction is necessary in patients with large tracheal defects. Previously, artificial tracheae made of polypropylene and collagen sponge have been used clinically by our group. As a basic research aimed at promoting epithelialization for infection defense, we transplanted cell sheets of human induced pluripotent stem cell (hiPSC)-derived airway epithelial cells (iAECs) with artificial tracheae into tracheal defects of rats and confirmed their engraftment.

Background: Doublesex and mab-3 related transcription factor (DMRT) 1, commonly found in all vertebrates, regulates the transcription of genes involved in the masculinization and maintenance of gonadal somatic cells and/or germline cell development. DMRT1 has a DNA-binding domain called the DM domain and a transcription regulatory region. Unlike the former, there is little knowledge about the latter transcription regulatory region.

In certain tetraploid species resulting from interspecific hybridization, one parent's subgenome is known to selectively undergo DNA loss. The molecular mechanisms behind this remain unclear. In our study, we compared the genomes of a standard diploid species with two allotetraploid species from the Xenopus genus, both possessing L (longer) and S (shorter) homoeologous subgenomes.

Human vocal folds (VFs), a pair of small, soft tissues in the larynx, have a layered mucosal structure with unique mechanical strength to support high-level tissue deformation by phonation. Severe pathological changes to VF have causes including surgery, trauma, age-related atrophy, and radiation, and lead to partial or complete communication loss and difficulty in breathing and swallowing. VF glottal insufficiency requires injectable VF biomaterials such as hyaluronan, calcium hydroxyapatite, and autologous fat to augment VF functions.

  • Memory T cells are diverse and change over time, influenced by factors such as cytokines and antigen exposure, complicating the study of aging's effects on them.
  • A study on Epstein-Barr virus (EBV) showed that memory CD8 T cells from young adults (<40 years) exhibit specific differentiation patterns, whereas older adults (>65 years) show distinct aging paths with diminished adaptive features but increased innate characteristics.
  • Surprisingly, despite losing naïve/stem-like EBV-specific T cells with age, the overall diversity of EBV-specific memory T cells either remained stable or improved, providing insights for better vaccination and T cell therapy strategies.

Inhibiting the cytotoxic T-lymphocyte-associated protein-4 (CTLA-4)-mediated immune checkpoint system using an anti-CTLA-4 antibody (Ab) can suppress the growth of various cancers, but the detailed mechanisms are unclear. In this study, we established a monoclonal hepatocellular carcinoma cell line (Hepa1-6 #12) and analyzed the mechanisms associated with anti-CTLA-4 Ab treatment. Depletion of CD4+ T cells, but not CD8+ T cells, prevented anti-CTLA-4 Ab-mediated anti-tumor effects, suggesting dependence on CD4+ T cells.

  • Congenital portosystemic shunts (CPSSs) vary in phenotype and pose challenges in determining the best treatment and prognosis, with a study analyzing 122 cases in Japan from 2000-2019 to gather insights.
  • The majority of patients (61.5%) had portal vein (PV) visibility on imaging; however, complications like hyperammonemia and liver masses were notably more prevalent in those without PV visualization, indicating a link between imaging results and health outcomes.
  • While most patients received treatment, including surgical options, those without PV visualization were more likely to experience complications, suggesting the need for early intervention even in asymptomatic cases.

Constructing antifouling surfaces is a crucial technique for optimizing the performance of devices such as water treatment membranes and medical devices in practical environments. These surfaces are achieved by modification with hydrophilic polymers. Notably, zwitterionic (ZI) polymers have attracted considerable interest because of their ability to form a robust hydration layer and inhibit the adsorption of foulants.

The treatment of long-tracheal lesion is difficult because there are currently no viable grafts for tracheal replacement. To solve this problem, we have developed an autologous Tissue-Engineered Trachea (aTET), which is made up of collagenous tissues and cartilage-like structures derived from rat chondrocytes. This graft induced successful long-term survival in a small-animal experiment in our previous study.

Article Synopsis
  • The study evaluated the differences between pancreatic ductal adenocarcinoma (PDAC) located in the head (Ph) versus the body/tail (Pbt) in patients who received neoadjuvant chemoradiotherapy (NACRT) before surgery.
  • It included 107 patients who underwent surgery after NACRT, finding that the Ph group had lower albumin levels and worse immune markers compared to the Pbt group, which had higher rates of certain types of cancer spread.
  • Although the overall and recurrence-free survival rates were similar for both groups, the Pbt group had a significantly higher risk of recurrence via peritoneal dissemination.
