Background: The role of cyclic guanosine 3',5'-monophosphate (cGMP) after acute myocardial infarction (AMI) is not well understood despite its significance as a second messenger of natriuretic peptides (NPs) in cardiovascular disease. We investigated the association between the NP-cGMP cascade and left ventricular reverse remodelling (LVRR) in anterior AMI.

Methods: 67 patients with their first anterior AMI (median age, 64 years; male, 76%) underwent prospective evaluation of plasma concentrations of the molecular forms of A-type and B-type natriuretic peptide (BNP) and cGMP from immediately after primary percutaneous coronary intervention (PPCI) to 10 months post-AMI.

The presence of redox-active molecules containing catenated sulfur atoms (supersulfides) in living organisms has led to a review of the concepts of redox biology and its translational strategy. Glutathione (GSH) is the body's primary detoxifier and antioxidant, and its oxidized form (GSSG) has been considered as a marker of oxidative status. However, we report that GSSG, but not reduced GSH, prevents ischemic supersulfide catabolism-associated heart failure in male mice by electrophilic modification of dynamin-related protein (Drp1).

Background: Protein expression analysis of isolated brain microvessels provides valuable insights into the function of the blood-brain barrier (BBB). However, isolation of brain microvessels from human brain tissue, particularly in small quantities, poses significant challenges. This study presents a method for isolating brain microvessels from a small amount of frozen human brain tissue, adapting techniques from an established mouse brain capillary isolation method.

Article Synopsis
  • The study examines the link between oral microbiome diversity and mortality rates from all causes, cardiovascular disease (CVD), and non-CVD, highlighting gaps in knowledge regarding sex and racial/ethnic variations.
  • Using data from the National Health and Nutrition Examination Survey (NHANES), researchers analyzed oral rinse samples from 8,199 adults and tracked mortality over about 9 years to assess the impact of microbial diversity on health outcomes.
  • Findings indicate that higher oral microbiome diversity is associated with lower mortality rates across groups, particularly among Mexican Americans and Non-Hispanic Whites, but potential mediators like obesity or diabetes did not influence this relationship.

Impaired hepatic and renal function influence Alzheimer's disease (AD) progression; however, whether AD progression affects these important organ functions remains unclear. Here, we investigated the impact of AD progression, characterized by brain amyloid-beta (Aβ) accumulation, on liver and kidney function of App (APP-KI) mice using quantitative proteomics. SWATH-based quantitative proteomics revealed changes in mitochondrial, drug metabolism, and pharmacokinetic-related proteins in mouse liver and kidneys during the early (2-month-old) and intermediate (5-month-old) stages of Aβ accumulation.

Background: Video-assisted thoracoscopic surgery (VATS) is considered useful for the treatment of parapneumonic empyema in children. However, thoracoscopic management of empyema due to esophageal leakage as an operative complication has not been well described in the literature.

Case Presentation: We successfully decorticated severe empyema using uniport VATS in 2 children (a 2-year-old boy who suffered esophageal perforation after laparoscopic anti-reflux surgery, and a 7-month-old girl who had anastomotic leakage after thoracoscopic repair of esophageal atresia).

Article Synopsis
  • The study investigated how the CHADS-VASc score and prior oral anticoagulant (OAC) use affect endovascular treatment (EVT) outcomes in acute ischemic stroke patients with atrial fibrillation (AF).
  • Out of 6984 patients analyzed, a significant portion had high CHADS-VASc scores, which correlated with increased risks of complications and decreased independence post-treatment; however, prior use of direct oral anticoagulants (DOACs) improved some outcomes for high-risk patients.
  • Findings suggest that using DOACs in high- and selected moderate-risk AF patients prior to EVT can enhance treatment success, indicating potential benefits for preventive DOAC therapies.

Article Synopsis
  • Researchers identified AMIGO2 as a key driver gene in liver metastasis and poor outcomes for colorectal cancer (CRC) patients.
  • The study explored how AMIGO2 promotes epithelial-mesenchymal transition (EMT), which enhances the invasion potential of CRC cells, primarily through the activation of the TGFβ/Smad signaling pathway.
  • Findings indicate that AMIGO2 is significantly expressed in the invasive front of tumors and its nuclear presence is linked to EMT marker expression, suggesting it could be targeted for therapies aiming to inhibit metastasis in CRC.

Ice formation is a critical challenge across multiple fields, from industrial applications to biological preservation. Inspired by natural antifreeze proteins, we designed and synthesized a new class of small-molecule antifreezes based on α-helical -terphenyl scaffolds with guanidine side chains. These -terphenyl guanidines , among the smallest molecules that mimic α-helical structures, exhibit potent ice recrystallization inhibition (IRI) activity, similar to that of existing large α-helical antifreeze compounds.
