Article Synopsis
  • A study in Japan compared two treatment intervals for nivolumab (480 mg every 4 weeks vs. 240 mg every 2 weeks) to evaluate their efficacy, safety, and economic impact on cancer patients, as prior real-world data was lacking.
  • The analysis of 126 patients showed no significant differences in overall survival or adverse event rates between the two groups, indicating similar effectiveness and safety.
  • However, the 4-week treatment interval had lower medical costs (excluding drug costs) than the 2-week interval, suggesting a potential economic advantage for using the longer treatment period.

To computationally investigate the recent experimental finding such that extracellular ATP release caused by exogeneous mechanical forces promote wound closure, we introduce a mathematical model, the Cellular Potts Model (CPM), which is a popular discretized model on a lattice, where the movement of a "cell" is determined by a Monte Carlo procedure. In the experiment, it was observed that there is mechanosensitive ATP release from the leading cells facing the wound gap and the subsequent extracellular Ca influx. To model these phenomena, the Reaction-Diffusion equations for extracellular ATP and intracellular Ca concentrations are adopted and combined with CPM, where we also add a polarity term because the cell migration is enhanced in the case of ATP release.

Background: Bone metastases are frequently observed in advanced cancer, and bone modifying agents are used to prevent or treat skeletal-related events. Zoledronic acid is contraindicated in patients with severe renal impairment (Ccr < 30 mL/min), but it is not completely known whether denosumab can be used in them. We aimed to determine the association between renal function and hypocalcemia development during denosumab treatment.
