Synthesis of ribozyme against vascular endothelial growth factor165 and its biological activity in vitro.

Aim: To investigate the designation, synthesis and biological activity of against vascular endothelial growth factor165 (VEGF165) ribozyme.

Methods: The ribozyme against VEGF165 was designed with computer. The transcriptional vector was constructed which included the anti-VEGF165 ribozyme and 5', 3' self-splicing ribozymes.

[Effect of anti-VEGF₁₆₅ ribozyme on biological behaviors of human lung adenocarcinoma cell].

Background: To investigate the biological effects of anti VEGF₁₆₅ ribozyme on human lung adenocarcinoma cell.

Methods: Hammerhead ribozyme (VRz) against VEGF₁₆₅ gene transcripts (site 212) and its paired mutant ribozyme (mVRz) were designed and synthesized, and the cleavage activity of the ribozymes on target RNA in a cell-free system was observed. The replication-incompetent adenovirus-mediated eukaryotic expression vectors (rpAdVRz) containing VRz and mVRz gene were constructed and identified.

Computer Analysis of the Cleavage Reaction of Hammerhead Ribozyme.

We have analyzed various in vitro cleavage reactions of hammerhead ribozyme by a computer program. Three energy changes: deltaE(s'), deltaE(r'), and deltaE, were calculated. deltaE(s') was the hypothetical energy requirement for the substrate to be activated from the most stable state to the relaxed state.
