Article Synopsis
  • SK3 channels are potassium channels that contribute to cancer aggressiveness, and their regulation by endogenous ether lipids was previously unknown.
  • Experiments showed that inhibiting key enzymes involved in ether lipid synthesis reduced SK3 expression and decreased cancer cell behaviors like migration and invasion through increased levels of specific micro RNAs.
  • This study is the first to demonstrate that ether lipids influence tumor aggressiveness by regulating ion channels, suggesting potential new therapeutic strategies for cancer treatment.

Anti-PD1 immunotherapy, as a single agent or in combination with standard chemotherapies, has significantly improved the outcome of many patients with cancers. However, resistance to anti-PD1 antibodies often decreases the long-term therapeutic benefits. Despite this observation in clinical practice, the molecular mechanisms associated with resistance to anti-PD1 antibody therapy have not yet been elucidated.
