We compared the effects of resveratrol and rosiglitazone, alone and in combination, on indices of fatty acid re-esterification in cultured adipose tissue from obese participants (n = 17) undergoing gastric bypass. Rosiglitazone induced PDK4 and PEPCK gene expression to a greater extent than resveratrol. Co-treatment with both compounds induced PDK4 and PEPCK expression in parallel with reductions in the fatty acid to glycerol ratio.

Adipose tissue plays a key role in the development of type-2 diabetes via the secretion of adipokines. The current study investigated if secretion media derived from intact visceral (VAT) and subcutaneous (SAT) adipose tissues from extremely obese men and women differently suppressed insulin signaling in human skeletal myotubes derived from a healthy, non-diabetic male and female donor, respectively. Adipose tissue samples were collected from men and women during laparoscopic bariatric surgery.

Objective: Polyunsaturated fatty acids (PUFAs) regulate fatty acid desaturase (FADS1, FADS2) expression in the liver; however, it is unknown whether PUFAs regulate FADS in adipocytes. This is important to study considering reports that link altered desaturase activity with adipose tissue PUFA profiles, body weight, and whole-body glucose homeostasis. Therefore, the present study aimed to determine the direct effects of PUFAs on FADS expression in differentiated 3T3-L1 adipocytes.
