PTPMT1 was the first protein tyrosine phosphatase found localized to the mitochondria, but its biological function was unknown. Herein, we demonstrate that whole body deletion of Ptpmt1 in mice leads to embryonic lethality, suggesting an indispensable role for PTPMT1 during development. Ptpmt1 deficiency in mouse embryonic fibroblasts compromises mitochondrial respiration and results in abnormal mitochondrial morphology.

Leishmania parasites have evolved sophisticated mechanisms to subvert macrophage immune responses by altering the host cell signal transduction machinery, including inhibition of JAK/STAT signalling and other transcription factors such as AP-1, CREB and NF-κB. AP-1 regulates pro-inflammatory cytokines, chemokines and nitric oxide production. Herein we show that upon Leishmania infection, AP-1 activity within host cells is abolished and correlates with lower expression of 5 of the 7 AP-1 subunits.

Optimal scaffold characteristics are essential for the therapeutic application of engineered tissues. Hydraulic permeability (k) affects many properties of collagen gels, such as mechanical properties, cell-scaffold interactions within three dimensions (3D), oxygen flow and nutrient diffusion. However, the cellular response to 3D gel scaffolds of defined k values has not been investigated.

Flash ladar systems are compact devices with high frame rates that hold promise for robotics applications, but these devices suffer from poor spatial resolution. This work develops a wavelet preprocessing stage to enhance registration of multiple frames and applies super-resolution to improve the resolution of flash ladar range imagery. The triangle orientation discrimination methodology was used for a subjective evaluation of the effectiveness of super-resolution for flash ladar.

Information from experts, or "key informants," is often used when estimating the prevalence of a disease, the numbers in particular risk groups, or the frequency of particular risk behaviors. This study aimed to better understand and describe the usefulness of key informants in informing an area such as injection drug use, where the populations are often marginalized and difficult to identify and the illnesses (HIV and hepatitis C virus) associated with the risk behavior can lead to discrimination by the general community. Our study results highlight the limitations of relying upon key informant information alone to provide specific information or accurate data about ethnic Vietnamese injection drug users.

Extensive X-ray crystallographic studies carried out on the catalytic-subunit of protein kinase A (PKA-C) enabled the atomic characterization of inhibitor and/or substrate peptide analogues trapped at its active site. Yet, the structural and dynamic transitions of these peptides from the free to the bound state are missing. These conformational transitions are central to understanding molecular recognition and the enzymatic cycle.

Three cohorts of juvenile and subadult Chinook salmon Oncorhynchus tshawytscha received multiple treatments with macrolide antibiotics for bacterial kidney disease (BKD) during rearing in a captive broodstock program. A total of 77 mortalities among the cohorts were screened for Renibacterium salmoninarum, the etiologic agent of BKD, by agar culture from kidney, and isolates from 7 fish were suitable for growth testing in the presence of macrolide antibiotics. The minimum inhibitory concentration (MIC) of erythromycin and azithromycin was determined by a modification of the standardized broth assay using defined medium.

Trophoblast expression of the non-classical MHC, HLA-G, is considered essential for feto-maternal immune tolerance and successful placentation in pregnancy. The HLA-G 14bp polymorphism in the 3'-untranslated region (UTR) of the HLA-G gene has been reported to be associated with development of pre-eclampsia (PE). In this study, maternal (peripheral blood, n=54) and fetal (cord blood, n=57) HLA-G 14bp genotypes have been determined by PCR in pre-eclamptic and normal pregnancies.

The hepatitis C virus (HCV) affects over 170 million people worldwide. In Australia, over 225,000 people have been diagnosed with HCV infection with 13,000 infections reported annually; 90% are attributed to injecting drug use. Burnet Institute (BI) researchers have been studying the HCV epidemic since the virus was identified in 1989 including community based cohort studies (1990-1995), numerous studies involving Vietnamese-Australian people who inject drugs (PWID) (1996-2004) and social network studies (2000-2002, 2005-2007).

Despite current injecting drug users (IDUs) being the major risk group for new hepatitis C virus (HCV) infections in most countries, they constitute a small minority of study populations in almost all studies of acute HCV infection treatment. The Australian Trial in Acute Hepatitis C (ATAHC) is examining natural history and treatment efficacy among predominantly IDU-acquired acute HCV. Recruitment is through an Australian network of primary and tertiary care sites.

The alpha2A and alpha2C adrenergic receptor (AR) subtypes mediate antinociception when activated by the endogenous ligand norepinephrine. These receptors also produce antinociceptive synergy when activated concurrently with opioid receptor activation. The involvement of the opioid receptors in the mechanisms governing transcutaneous electrical nerve stimulation (TENS) has been well described.

Seizures in cortical dysplasia (CD) could be from cytomegalic neurons and balloon cells acting as epileptic 'pacemakers', or abnormal neurotransmission. This study examined these hypotheses using in vitro electrophysiological techniques to determine intrinsic membrane properties and spontaneous glutamatergic and GABAergic synaptic activity for normal-pyramidal neurons, cytomegalic neurons and balloon cells from 67 neocortical sites originating from 43 CD patients (ages 0.2-14 years).

Materials presenting a stable and reversible switch of physical properties in the solid state are of major interest either for fundamental interests or potential industrial applications. In this context, the design of metal complexes showing a light-induced crossover from one spin state to another, leading to a major change of magnetic and optical properties, is probably one of the most appealing challenges. The so-denoted spin-crossover materials undergo, in some cases, a reversible photoswitch between two magnetic states, but, unfortunately, lifetimes of the photomagnetic states for compounds known so far are long enough only at low temperatures; this prohibits any applications.

Huntington's disease (HD) is characterized by loss of striatal gamma-aminobutyric acid (GABA)ergic medium-sized spiny projection neurons (MSSNs), whereas some classes of striatal interneurons are relatively spared. Striatal interneurons provide most of the inhibitory synaptic input to MSSNs and use GABA as their neurotransmitter. We reported previously alterations in glutamatergic synaptic activity in the R6/2 and R6/1 mouse models of HD.
