The primary cilium is a microtubule-based organelle that cycles through assembly and disassembly. In many cell types, formation of the cilium is initiated by recruitment of ciliary vesicles to the distal appendage of the mother centriole. However, the distal appendage mechanism that directly captures ciliary vesicles is yet to be identified.

Current lithium batteries experience significant performance degradation under extreme temperature conditions, both high and low. Traditional wide-temperature electrolyte designs typically addressed these challenges by manipulating the solvation sheath and selecting solvents with extreme melting/boiling points. However, these solvent-mediated solutions, while effective at one temperature extreme, invariably fail at the opposite end due to the inherent difficulties in maintaining solvent stability across wide temperatures.

Elucidating the relationships between a class I peptide antigen, a CD8 T cell receptor (TCR) specific to that antigen, and the T cell phenotype that emerges following antigen stimulation, remains a mostly unsolved problem, largely due to the lack of large data sets that can be mined to resolve such relationships. Here, we describe Antigen-TCR Pairing and Multiomic Analysis of T-cells (APMAT), an integrated experimental-computational framework designed for the high-throughput capture and analysis of CD8 T cells, with paired antigen, TCR sequence, and single-cell transcriptome. Starting with 951 putative antigens representing a comprehensive survey of the SARS-CoV-2 viral proteome, we utilize APMAT for the capture and single cell analysis of CD8 T cells from 62 HLA A*02:01 COVID-19 participants.

A growing number of therapies are being developed to target the cell cycle machinery for the treatment of cancer and other human diseases. Consequently, a greater understanding of the factors regulating cell cycle progression becomes essential to help enhance the response to these new therapies. Here, using data from the Cancer Dependency Map, we identified the poorly-studied factor FAM53C as a new regulator of cell cycle progression.

While spatial transcriptomics offer valuable insights into gene expression patterns within the spatial context of tissue, many technologies do not have a single-cell resolution. Here, we present SMART, a marker gene-assisted deconvolution method that simultaneously infers the cell type-specific gene expression profile and the cellular composition at each spot. Using multiple datasets, we show that SMART outperforms the existing methods in realistic settings.

In Singapore's rapidly ageing population where the demand for tailored healthcare services is increasing, but is confronted with a shortage of geriatric medicine trained physicians, innovative and sustainable ways to provide quality healthcare care for frail older adults need to be explored. Recognising, developing, growing and establishing the role of geriatric Advanced Practice Nurses (APNs) in Singapore, not just in clinical practice but in education and research as well, potentially offers one such solution to this problem. This narrative review, therefore, examines the available literature on how geriatric APNs can meet the healthcare needs of frail older adults in various local healthcare settings.

The study of olfaction in has greatly benefited from genetic reagents such as olfactory receptor mutant lines and GAL4 reporter lines. The CRISPR/Cas9 gene-editing system has been increasingly used to create null receptor mutants or replace coding regions with GAL4 reporters. To further expand this toolkit for manipulating fly olfactory receptor neurons (ORNs), we generated null alleles for 11 different olfactory receptors by using CRISPR/Cas9 to knock in LexA drivers, including multiple lines for receptors which have thus far lacked knock-in mutants.

Article Synopsis
  • The study examines sleep behaviors in individuals with Cornelia de Lange syndrome (CdLS) caused by variants in two genes, NIPBL and SMC1A, which are linked to developmental delays.
  • Caregivers of 31 individuals provided data showing that those with NIPBL variants exhibited more behavior regulation difficulties and repetitive behaviors compared to those with SMC1A variants.
  • The research highlights a strong connection between sleep disturbances and behavior regulation challenges, particularly in the NIPBL group, suggesting that sleep interventions may help manage behavioral issues differently depending on the genetic variant.
