Background: Muscarinic receptor agonism and positive allosteric modulation is a promising mechanism of action for treating psychosis, not present in most D2R-blocking antipsychotics. Xanomeline, an M1/M4-preferring agonist, has shown efficacy in late-stage clinical trials, with more compounds being investigated. Therefore, we aim to synthesize evidence on the preclinical efficacy of muscarinic receptor agonists and positive allosteric modulators in animal models of psychosis to provide unique insights and evidence-based information to guide drug development.

Phagocytosis by immunocompetent cells is a key role in the biological defense mechanism and is the starting point of the reaction that leads from innate to acquired immunity. Several studies have demonstrated that some lactic acid bacteria strains activate the innate and acquired immune systems of the host. However, further investigation of the mechanism and improvement of usefulness is needed because the effect differs depending on the type and strain of lactic acid bacteria.

Article Synopsis
  • - The study investigates how SARS-CoV-2 buds and transports its virions from the ERGIC to the cell surface, revealing that the vesicles involved have a protein coat known as coatomer complex I (COPI).
  • - Researchers observed that during infection, the distribution of COPI and the ERGIC changed, suggesting they play a role in the virus's replication process.
  • - Depleting a key COPI component, COPB2, confined the SARS-CoV-2 virions within the ERGIC and significantly reduced viral release, indicating that targeting COPI could be a potential strategy for developing antiviral treatments.

  • Idiopathic intracranial hypertension (IIH) is a condition with high brain pressure that can severely affect vision, and its cause is often unknown.
  • Recent studies suggest that venous sinus stenting (VSS) may be an effective treatment, as demonstrated by two young women who experienced quick relief from visual problems after the procedure.
  • Both patients showed elevated cerebrospinal fluid pressure and narrowed transverse sinuses, and after using stents to widen these sinuses, their symptoms and papilledema improved significantly right away.

Macaque restricts hepatitis B virus (HBV) infection because its receptor homologue, NTCP (mNTCP), cannot bind preS1 on viral surface. To reveal how mNTCP loses the viral receptor function, we here solve the cryo-electron microscopy structure of mNTCP. Superposing on the human NTCP (hNTCP)-preS1 complex structure shows that Arg158 of mNTCP causes steric clash to prevent preS1 from embedding onto the bile acid tunnel of NTCP.

Phospholipids are asymmetrically distributed in the plasma membrane (PM), with phosphatidylcholine and sphingomyelin abundant in the outer leaflet. However, the mechanisms by which their distribution is regulated remain unclear. Here, we show that transmembrane protein 63B (TMEM63B) functions as a membrane structure-responsive lipid scramblase localized at the PM and lysosomes, activating bidirectional lipid translocation upon changes in membrane curvature and thickness.

Objectives: Appropriate sedative and analgesic selection is essential to reduce patient discomfort and body movement to safely conduct endoscopic ultrasonography (EUS). However, few cases have examined sedation with propofol in EUS, and few studies the need for combined analgesia. In this study, we retrospectively evaluated the usefulness and safety of propofol without analgesics for sedation in biliopancreatic observational EUS.

Article Synopsis
  • The study focused on comparing the effectiveness of Pipet Curet cytology (PCC) and Pipet Curet biopsy (PCB) for diagnosing endometrial endometrioid carcinoma (EEC) in 77 patients, categorized based on the Federation of Gynecology and Obstetrics (FIGO) grades.
  • Results showed that the diagnostic sensitivity was significantly higher for advanced G2/3 EEC cases than for G1 EECs across all methods, indicating that more aggressive cancer types yield easier detection.
  • The combination of both PCC and PCB improved diagnostic accuracy, suggesting that using these methods together can reduce false negatives and enhance EEC diagnosis.

Background: Severe psychomotor agitation and aggression often require immediate pharmacological intervention, but clear evidence-based recommendations for choosing among the multiple options are lacking. To address this gap, we plan a systematic review and individual-participant-data network meta-analysis to investigate their comparative effectiveness in real-world emergency settings with increased precision.

Methods: We will include randomized controlled trials investigating intramuscular or intravenous pharmacological interventions, as monotherapy or in combination, in adults with severe psychomotor agitation irrespective of the underlying diagnosis and requiring rapid tranquilization in general or psychiatric emergency settings.
