Energy metabolism of chimeric antigen receptor-T cells (CAR-T) activation in humans remains unexplored. As a glycolytic activity surrogate, we investigated the dynamics of peripheral blood (PB) lactate in the first weeks post-CAR-T infusion. In 17 patients treated with CD28 harbording anti-CD19 CAR-T for relapsed/refractory non-Hodgkin lymphomas, PB lactate levels increased following CAR-T infusion.

Article Synopsis
  • Donor cell engraftment is necessary for successful stem cell transplantation, but the cellular interactions in bone marrow during this process have not been well understood.
  • Researchers used mass cytometry and CITEseq to analyze bone marrow cells three months after transplantation in patients with leukemia, finding significant changes in myeloid and B-cell progenitors, indicating a shift towards inflammation-driven hematopoiesis.
  • The study concluded that bone marrow engraftment is marked by emergency hematopoiesis and an inflammatory response, particularly in patients who experienced graft-versus-host disease (GVHD).

Article Synopsis
  • Bispecific antibodies (BsAbs) are used to treat relapsed or refractory multiple myeloma, but frequent infections were noted during clinical trials, indicating a need for further real-world data on infection risks and outcomes.
  • A multicenter retrospective study involving 229 patients in France from December 2020 to February 2023 found a high incidence of infections, with 234 recorded cases, primarily affecting the respiratory system, and a hospitalization rate of 56%.
  • The study identified corticosteroids used for managing cytokine release syndrome as a risk factor for infections, while using GPRC5D-targeting BsAbs and antibacterial prophylaxis was associated with a reduced risk.
