Article Synopsis
  • This study explored how varying levels of liver dysfunction affect the pharmacokinetics and safety of dolasetron, a medication with both kidney and liver elimination pathways.
  • Seventeen adults were divided into three groups: healthy volunteers, those with mild liver impairment, and those with moderate to severe liver impairment, to compare their responses to single doses of dolasetron.
  • Findings indicated no significant differences in drug absorption or clearance among the groups following intravenous administration, although oral administration showed more notable effects in patients with severe liver impairment, with longer half-lives and altered metabolite profiles, while overall the drug was well tolerated across all groups.

The effect of the timing of a standard meal relative to a single oral dose of 200 mg ibopamine, on the appearance of its pharmacologically active metabolite, epinine, in plasma was investigated in a randomised crossover study in 12 healthy volunteers. After a 12 h fast, ibopamine was administered either in the fasting state (no meal), or 1 h before, 0.5 h before, immediately after, 2 h after or 3 h after a standardised meal.
