Objective: To describe the comprehensive activities implemented in the Bolivarian Republic of Venezuela to achieve reverification of measles elimination by 2023.

Methods: Descriptive study of the procedures implemented to achieve reverification of measles elimination by 2023 according to the components set forth in the Regional Framework for the Monitoring and Re-verification of Measles, Rubella, and Congenital Rubella Syndrome Elimination in the Americas, published in 2022.

Results: Due to a measles outbreak that began in epidemiological week (EW) 26 of 2017, Venezuela lost the elimination status which had been conferred in 2016 by the Expert Committee.

Objective: Describe the challenges, strategies and lessons learned in Venezuela during the measles outbreak of 2017-2019, and discuss the mechanisms employed for its containment, which led to the interruption of endemic transmission.

Methods: Descriptive study of the actions taken by Venezuela to interrupt the outbreak.

Results: When the outbreak was confirmed, the Venezuelan government, with technical cooperation from the Pan American Health Organization, activated a plan to interrupt measles transmission.

The imidazo[1,5-a]quinoline scaffold of central benzodiazepine receptor (CBR) ligands was used as the pharmacophore in the design of bivalent ligands bearing spacers showing variable length and different physicochemical features. The newly designed compounds were synthesized along with the corresponding reference monovalent compounds bearing the corresponding spacers terminated with a tert-butoxycarbonyl group. The novel compounds were tested in binding assays with different CBR preparations such as the cerebral cortex from male CD-1 albino mice or the human recombinant α1β3γ2 and α2β3γ2 γ-aminobutyric acid type A receptors (GABARs) stably expressed in mouse L(tk-) cells.

There is a critical need for safer and better-tolerated alternatives to address the current limitations of antidepressant treatments for major depressive disorder. Recently, drugs targeting the GABA system via α5-containing GABA receptors (α5-GABAR) as negative allosteric modulators (α5-NAMs) have shown promise in alleviating stress-related behaviors in preclinical studies, suggesting that α5-NAMs may have translational relevance as novel antidepressant medications. Here, we evaluated the efficacy of Basmisanil, an α5-NAM that has been evaluated in Phase 2 clinical studies as a cognitive enhancer, in a battery of behavioral tests relevant to coping strategies, motivation, and aversion in male mice, along with plasma and brain pharmacokinetic measurements.
