A new gas analytical technique for infrared laser spectroscopy combined with differential pressure measurement (IR-DP) is developed. The basic idea of this technique is that the absorption process of molecules by laser irradiation is monitored as the pressure enhancement in a gas cell by use of a differential pressure gauge. The system is composed of a tunable IR laser system, sample cell, differential pressure gauge, and data accumulation system.

Article Synopsis
  • Pulmonary tumor thrombotic microangiopathy (PTTM) is a serious and rare complication linked to malignant tumors, leading to severe pulmonary hypertension and often resulting in respiratory failure.
  • In a reported case, a 70-year-old man with gastric cancer was diagnosed with PTTM after dying during chemotherapy; the autopsy showed specific signs of this condition.
  • The study highlighted the presence of vascular growth factors in the cancer cells and suggested that versican (VCAN), an important protein in blood vessel abnormalities, plays a role in the development of PTTM.

We examined the conformation of benzo-15-crown-5 (B15C5) and benzo-18-crown-6 (B18C6) complexes with ammonium ions, NH, CHNH (MeNH), CHCHNH (EtNH), and CHCHCHNH (PrNH), using cold UV and IR spectroscopy in the gas phase. We measured the UV photodissociation (UVPD) spectra of the ammonium complexes and compared them with those of the K(B15C5) and K(B18C6) complexes in order to identify the conformation on the basis of the band position. The number of possible conformations for the ammonium complexes of B15C5 is limited compared with alkali metal ions with similar ionic radii.

Shati/Nat8l was identified as an upregulated molecule in the nucleus accumbens (NAc) of mice following repeated methamphetamine administration. Region-specific roles of this molecule are associated with psychiatric disorders. In the present study, we examined the importance of Shati/Nat8l in the hippocampus because of its high expression in this region.

Introduction: In gene therapy with adeno-associated virus (AAV) vectors for diseases of the central nervous system, the vectors can be administered into blood vessels, cerebrospinal fluid space, or the brain parenchyma. When gene transfer to a large area of the brain is required, the first two methods are used, but for diseases in which local gene transfer is expected to be effective, vectors are administered directly into the brain parenchyma.

Areas Covered: Strategies for intraparenchymal vector delivery in gene therapy for Parkinson's disease, aromatic l-amino acid decarboxylase (AADC) deficiency, and epilepsy are reviewed.

Idling brain activity has been proposed to facilitate inference, insight, and innovative problem-solving. However, it remains unclear how and when the idling brain can create novel ideas. Here, we show that cortical offline activity is both necessary and sufficient for building unlearned inferential knowledge from previously acquired information.

Radical cations of dimeric clusters of carbon dioxide/disulfide, [(CX)] (X = O and S), form strong intracluster bonds through charge resonance (CR) interactions. We herein performed electronic photodissociation spectroscopy of [(CX)] while regulating the temperature under ambient and cryogenic conditions using a quadrupole ion trap. Both ions exhibited broad band absorption in the near-infrared-visible light region; it is called the "CR band", as a measure of the strength of the CR interaction.

Article Synopsis
  • - Recent Marburg virus disease outbreaks in Guinea, Ghana, Equatorial Guinea, and Tanzania highlight increased spillover risks from natural host animals, signaling the need for better diagnostic tools.
  • - Researchers developed a rapid immunochromatography (IC) assay using monoclonal antibodies, which effectively detects Marburg virus (MARV) and Ravn virus (RAVV) in infected samples.
  • - The IC assay showed reliability in identifying viral antigens in serum from infected nonhuman primates, suggesting it could serve as a fast diagnostic method for Marburg virus disease in clinical settings.

Depression is a frequent and serious illness, and stress is considered the main risk factor for its onset. First-line antidepressants increase serotonin (5-hydroxytryptamine; 5-HT) levels in the brain. We previously reported that an -acetyltransferase, Shati/Nat8l, is upregulated in the dorsal striatum (dSTR) of stress-susceptible mice exposed to repeated social defeat stress (RSDS) and that dSTR Shati/Nat8l overexpression in mice (dSTR-Shati OE) induces stress vulnerability and local reduction in 5-HT content.

  • * Intravenous injection of AAV.GT5 resulted in similar liver transduction in both humanized mice and macaques, but it had a low recovery and short half-life due to its positive charge affecting blood interactions.
  • * Enhanced liver transduction was achieved by administering AAV.GT5 directly into the liver, and unlike AAV-Spark100, AAV.GT5 showed a lower tendency to generate neutralizing antibodies, suggesting it

The inner ear is a primary lesion in sensorineural hearing loss and has been a target in gene therapy. The efficacy of gene therapy depends on achieving sufficient levels of transduction at a safe vector dose. Vectors derived from various adeno-associated viruses (AAVs) are predominantly used to deliver therapeutic genes to inner ear cells.

CDC45-MCM2-7-GINS (CMG) helicase assembly is the central event in eukaryotic replication initiation. In yeast, a multi-subunit "pre-loading complex" (pre-LC) accompanies GINS to chromatin-bound MCM2-7, leading to CMG formation. Here, we report that DONSON, a metazoan protein mutated in microcephalic primordial dwarfism, is required for CMG assembly in vertebrates.
