Modulation of hippocampal synaptic plasticity by glucocorticoids has been attracting much attention, due to its importance in stress responses. Dendritic spines are essential for memory storage processes. Here, we investigated the effect of dexamethasone (DEX), a specific agonist of glucocorticoid receptor (GR), on density and morphology of dendritic spines in adult male rat hippocampus by imaging of Lucifer Yellow-injected spines in slices.

Formation of morphologically similar globular aggregates from diverse aggregation-prone proteins in mammalian cells.

Huntington's disease is a progressive neurodegenerative disorder caused by a polyglutamine repeat expansion in the first exon of the huntingtin (Htt) protein. N-terminal Htt peptides with polyglutamine tracts in the pathological range (51-122 glutamines) form high-molecular-weight protein aggregates with fibrillar morphology in vitro, and they form discrete inclusion bodies in a cell-culture model. However, in some studies, formation of discrete Htt inclusions does not correlate well with cell death.

GRIFFIN: a system for predicting GPCR-G-protein coupling selectivity using a support vector machine and a hidden Markov model.

We describe a novel system, GRIFFIN (G-protein and Receptor Interaction Feature Finding INstrument), that predicts G-protein coupled receptor (GPCR) and G-protein coupling selectivity based on a support vector machine (SVM) and a hidden Markov model (HMM) with high sensitivity and specificity. Based on our assumption that whole structural segments of ligands, GPCRs and G-proteins are essential to determine GPCR and G-protein coupling, various quantitative features were selected for ligands, GPCRs and G-protein complex structures, and those parameters that are the most effective in selecting G-protein type were used as feature vectors in the SVM. The main part of GRIFFIN includes a hierarchical SVM classifier using the feature vectors, which is useful for Class A GPCRs, the major family.

Acute repetitive transcranial magnetic stimulation reactivates dopaminergic system in lesion rats.

Repetitive transcranial magnetic stimulation (rTMS) offers potential benefit as a therapeutic treatment for neurological and psychiatric disorders. However, the mechanism underlying the therapeutic effects of rTMS is still unknown. In this study, we investigated the rescue effects of rTMS in the lesioned rats by administering the neurotoxin MPTP (l-methyl-4-phenyl-l,2,3,6-tetrahydropyridine).

[structure: see text] This paper describes the cyclotrimerization reaction of di(2-azulenyl)acetylenes (2a,b) catalyzed by Co2(CO)8 to produce hexa(2-azulenyl)benzene derivatives (1a,b). The cyclooligomerization of 2a and 2b utilizing CpCo(CO)2 as a catalyst produced (eta5-cyclopentadienyl)[tetra(2-azulenyl)cyclobutadiene]cobalt complexes (3a,b). The redox behavior of hexakis(6-octyl-2-azulenyl)benzene (1b), bis(6-octyl-2-azulenyl)acetylene (2b), and the cobalt complexes 3a and 3b along with 6-octyl-2-phenylazulene (19) was examined by cyclic voltammetry (CV).

Involvement of protein kinase PKN1 in G2/M delay caused by arsenite.

PKN1 is a serine/threonine protein kinase that has been reported to mediate cellular response to stress. We show here that in response to arsenite exposure, PKN1 kinase activity was stimulated, which was associated with increased binding of PKN1 to Cdc25C and delayed mitotic entry. A role for PKN1 in mediating arsenite-induced G(2)/M delay was supported by the finding that expression of a constitutively active form of PKN1 (PKN1AF3) in HeLa cells delayed the mitotic entry of cell cycle.

EWSR1 is fused to POU5F1 in a bone tumor with translocation t(6;22)(p21;q12).

POU5F1(OCT3/4) is a sequence-specific transcription factor that is essential for keeping germ cells and embryonic stem cells in an immature and pluripotent status. In this article, we report that POU5F1 was fused to EWSR1 in a case of undifferentiated sarcoma derived from pelvic bone with chromosomal translocation t(6;22)(p21;q12). The EWSR1-POU5F1 chimera consists of exons 1-6 of EWSR1 and exons 2-5 and a part of exon 1 of POU5F1.

Centrosome-targeting region of CG-NAP causes centrosome amplification by recruiting cyclin E-cdk2 complex.

Centrosome duplication occurs once per cell cycle and is thought to be triggered by cyclin E-cdk2. However, it is largely unknown how the duplication is regulated. Here, we found that the expression of the centrosome-targeting region of CG-NAP (centrosome and Golgi-localized PKN-associated protein), which we designate as CG-NAP/D, increased the number of centrosomes in Chinese hamster ovary (CHO)-K1 cells.

Calcineurin, a protein phosphatase required for Ca2+ signaling in many cell types, is a heterodimer composed of catalytic and regulatory subunits. Butyltin compounds (BTs) including mono-(MBT), di-(DBT) and tributyltin (TBT) were determined in livers of humans and wild terrestrial mammals, such as raccoon dogs (Nyctereutes procyonoids) and monkeys (Macaca fuscata) from Japan. In addition, 22 samples of plastic products were analyzed. BT residues were detected in all the liver samples of humans and raccoon dogs, with concentrations of <360 ng/g wet wt, whereas concentrations in the liver of monkeys were either less than the detection limit or were only in trace levels.

Acute urinary retention associated with fractured osteophyte.

We describe the case of a 40-year-old man whose disease was initially diagnosed as acute myelocytic leukemia. The patient achieved remission with chemotherapy, but relapsed shortly afterwards with an acute T-cell lymphoblastic leukemia. He died of intracranial bleeding.

A black-colored and well-circumscribed renal tumor in a 71-year-old woman is reported. The tumor was unique in that it was rich in vasculature and exclusively composed of perivascular epithelioid clear cells. Morphological features were reminiscent of conventional renal cell carcinoma (RCC).

In adult mammalian brain, occurrence of the synthesis of estradiol from endogenous cholesterol has been doubted because of the inability to detect dehydroepiandrosterone synthase, P45017alpha. In adult male rat hippocampal formation, significant localization was demonstrated for both cytochromes P45017alpha and P450 aromatase, in pyramidal neurons in the CA1-CA3 regions, as well as in the granule cells in the dentate gyrus, by means of immunohistochemical staining of slices. Only a weak immunoreaction of these P450s was observed in astrocytes and oligodendrocytes.

Except for white fat in male and female rats, tissue radioactivity concentrations 5 min after administration were 2 to 100 times the plasma values, evidence that the drug is widely distributed throughout the body.
