Quantitatively mapping enzyme sequence-catalysis landscapes remains a critical challenge in understanding enzyme function, evolution, and design. Here, we expand an emerging microfluidic platform to measure catalytic constants- and -for hundreds of diverse naturally occurring sequences and mutants of the model enzyme Adenylate Kinase (ADK). This enables us to dissect the sequence-catalysis landscape's topology, navigability, and mechanistic underpinnings, revealing distinct catalytic peaks organized by structural motifs.

Benzotriazoles (BZTs) and benzothiazoles (BTs) are high-production-volume chemicals utilized in many different commercial products and industrial processes, such as metal corrosion inhibitors, vulcanization accelerators, plastic-associated UV stabilizers, and pharmaceutical precursors. This study assessed age, gender, and temporal trends of BZTs and BTs in deidentified surplus pathology urine samples, pooled and stratified by age, gender, and sample collection year from a general Australian population (168 pools representing 16,800 individuals). Tolyltriazole (TTri), 5,6-dimethyl-1H-benzotriazole (DMBZT), 1,3-benzothiazole (BTH), 2-hydroxybenzothiazole (2-OH-BTH), and 2-aminobenzothiazole (2-amino-BTH) were detected in >50% of the pools.

Analyzing electroencephalogram (EEG) signals to detect the epileptic seizure status of a subject presents a challenge to existing technologies aimed at providing timely and efficient diagnosis. In this study, we aimed to detect interictal and ictal periods of epileptic seizures using a spiking neural network (SNN). Our proposed approach provides an online and real-time preliminary diagnosis of epileptic seizures and helps to detect possible pathological conditions.

This cross-cutting review focuses on the presence and impacts of per- and polyfluoroalkyl substances (PFAS) in the Arctic. Several PFAS undergo long-range transport via atmospheric (volatile polyfluorinated compounds) and oceanic pathways (perfluorinated alkyl acids, PFAAs), causing widespread contamination of the Arctic. Beyond targeting a few well-known PFAS, applying sum parameters, suspect and non-targeted screening are promising approaches to elucidate predominant sources, transport, and pathways of PFAS in the Arctic environment, wildlife, and humans, and establish their time-trends.

Aim: To develop a Theory of Change (ToC) pathway to facilitate the development of a multi-component intervention package supporting pressure Ulcer (PU) risk identification and management, in partnership with people with Long Term Neurological Conditions (LTNC) who self-manage care and live at home, their informal carers and PAs.

Methods: A participatory approach, with extensive input from those whose lives are the focus of the research, was used throughout the 4 interlinked work packages (WP): Iterative data analysis was undertaken with emerging findings from each WP informing subsequent stages of the study.

Findings: Overall, 74 participants contributed across the 4 WPs, incorporating 31 Service Users (SU), 8 carers, 9 Personal Assistants (PAs) and 26 professional stakeholders.

Ultraviolet (UV) absorbents and industrial antioxidants are two groups of plastic-derived contaminants of emerging environmental concern. However, their distribution and fate are poorly understood in Arctic wildlife. In the present study, 16 UV absorbents (10 benzotriazole UV stabilizers (BZT-UVs) and 6 organic UV filters (UVFs)) and 7 industrial antioxidants (6 aromatic secondary amines (Ar-SAs) and 2,6-di-tert-butylphenol (26DTBP)) were analyzed in the livers of thick-billed murre (Uria lomvia; n = 28), northern fulmar (Fulmarus glacialis; n = 4), black guillemot (Cepphus grylle; n = 11), polar bear (Ursus maritimus; n = 18), beluga whale (Delphinapterus leucas; n = 10), landlocked (n = 25) and sea-run (n = 10) Arctic char (Salvelinus alpinus) from the Canadian Arctic collected between 2017 and 2021.

Polycyclic aromatic hydrocarbons (PAHs), released from petrogenic, pyrogenic or diagenetic sources (degradation of wood materials), are of global concern due to their adverse effects, and potential for long-range transport. While dissolved PAHs have been frequently reported in the literature, there has been no consistent approach of sampling across water bodies. Germanium-on-Silicon (Ge-on-Si) avalanche photodiodes (APDs) are of considerable interest as low intensity light detectors for emerging applications. The Ge absorption layer detects light at wavelengths up to ≈ 1600 nm with the Si acting as an avalanche medium, providing high gain with low excess avalanche noise. Bottlenose dolphins () are keystone and sentinel species in the world's oceans. We studied correlations between per- and polyfluoroalkyl substances (PFAS) and their stress axis. We found that a winter of abnormally low snowfall and numerous dust storms from eolian processes acting on exposed landscapes (including a major 4-day dust storm while onsite in May 2014) caused a cascade of impacts on the physical, chemical, and ecological functioning of the largest lake by volume in the High Arctic (Lake Hazen; Nunavut, Canada). MODIS imagery revealed that dust deposited in snowpacks on the lake's ice acted as light-absorbing impurities (LAIs), reducing surface reflectance and increasing surface temperatures relative to normal snowpack years, causing early snowmelt and drainage of meltwaters into the lake. LAIs remaining on the ice surface melted into the ice, causing premature candling and one of the earliest ice-offs and longest ice-free seasons on record for Lake Hazen.

In the neocortex, neural activity is shaped by the interaction of excitatory and inhibitory neurons, defined by the organization of their synaptic connections. Although connections among excitatory pyramidal neurons are sparse and functionally tuned, inhibitory connectivity is thought to be dense and largely unstructured. By measuring in vivo visual responses and synaptic connectivity of parvalbumin-expressing (PV+) inhibitory cells in mouse primary visual cortex, we show that the synaptic weights of their connections to nearby pyramidal neurons are specifically tuned according to the similarity of the cells' responses.

Synchrotron X-ray imaging and spectroscopy techniques were used for studying changes during post-harvest storage of food grains. Three varieties (AAC Spitfire, CDC Defy, and AAC Stronghold) of the Canada Western Amber Durum (CWAD) wheat class were stored for five weeks at 17 % moisture content (wb).
