Objective: Cytotoxic agents are the cornerstone of treatment for patients with advanced intrahepatic cholangiocarcinoma (iCCA), despite heterogeneous benefit. We hypothesised that the pretreatment molecular profiles of diagnostic biopsies can predict patient benefit from chemotherapy and define molecular bases of innate chemoresistance.

Background: RNA modifications are important regulators of transcript activity and an increasingly emerging body of data suggests that the epitranscriptome and its associated enzymes are altered in human tumors.

Methods: Combining data mining and conventional experimental procedures, NSUN7 methylation and expression status was assessed in liver cancer cell lines and primary tumors. The clinical utility of serum alpha-fetoprotein (AFP) in patients with hepatocellular carcinoma (HCC) is widely recognised. However, a clear understanding of the mechanisms of AFP overexpression and the molecular traits of patients with AFP-high tumours are not known. In 6 studies, we found that advice is more idealistic than choice in decisions that trade off idealistic and pragmatic considerations. We propose that because advisers are more psychologically distant from the choosers' decision problem, they construe the dilemma at a higher construal level than do choosers (Trope & Liberman, 2003, 2010). Consequently, advisers are more influenced by idealistic considerations that are salient at a high-level construal, whereas choosers are more influenced by pragmatic considerations that are salient at a low-level construal.
