Background: Premenopausal women diagnosed with breast cancer often face aggressive chemotherapy resulting in infertility. Tamoxifen (TAM) is a selective estrogen receptor modulator that was previously suggested as a protective agent against chemotherapy-induced ovarian failure. In the current study, we examined mechanisms of the protective action of TAM in the ovaries of tumor-bearing rats treated with the chemotherapy drug cyclophosphamide (CPA).

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is a man-made chemical compound contaminating the environment. An exposure of organisms to TCDD results in numerous disorders. The main mechanism of TCDD action involves the induction of the aryl hydrocarbon receptor (AhR) pathway followed by the increase in the expression and activity of cytochrome P450 family 1 (CYP1) enzymes.

This review article contains a concise consideration of genetic and environmental risk factors for colorectal cancer. Known risk factors associated with colorectal cancer include familial and hereditary factors and lifestyle-related and ecological factors. Lifestyle factors are significant because of the potential for improving our understanding of the disease.

Background: 2,3,7,8-tetrachlorodibenzo--dioxin (TCDD) is a toxic man-made chemical, adversely affecting reproductive processes. The well-characterized canonical mechanism of TCDD action involves the activation of aryl hydrocarbon receptor (AhR) pathway, but AhR-independent mechanisms were also suggested. By applying RNA interference technology and Next Generation Sequencing (NGS) we aimed to identify genes involved in the mechanism of TCDD action in AhR knock-down porcine granulosa cells.

2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) is a toxic man-made chemical compound contaminating the environment and affecting human/animal health and reproduction. Intracellular TCDD action usually involves the activation of aryl hydrocarbon receptor (AhR). The aim of the current study was to examine TCDD-induced changes in the proteome of AhR-silenced porcine granulosa cells.

The 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) compound is an environmental chemical adversely affecting reproductive processes. Intracellular TCDD effects are mediated via aryl hydrocarbon receptor (AhR). The aim of the current study was to identify genes linking the AhR pathway with phenotypic consequences of TCDD action in granulosa cells of pigs.

Article Synopsis
  • The study identified and characterized 1,666 long non-coding RNAs (lncRNAs) in porcine granulosa cells, highlighting their structural differences compared to mRNAs.
  • TCDD exposure influenced the expression of 22 lncRNAs, suggesting these DELs may play roles in regulating gene expression related to xenobiotic responses and cell functions.
  • This research is the first to explore lncRNAs in porcine granulosa cells and their response to TCDD, paving the way for future studies on how these molecules impact ovarian biology under environmental stress.

Article Synopsis
  • TCDD is the most toxic and least biodegradable dioxin, while previous research indicated that pig CYP1A1 can hydroxylate another dioxin, DiCDD, but not TCDD.
  • Researchers used molecular modeling to study how pig CYP1B1 interacts with TCDD and DiCDD, finding that both dioxins stabilize in the enzyme’s active site but TCDD has a greater affinity, which may hinder its breakdown.
  • Despite both dioxins enhancing CYP1B1 gene expression and activity in pig granulosa cells, the structural constraints of CYP1B1 may limit TCDD's biodegradation effectiveness compared to DiCDD.

Article Synopsis
  • TCDD is a toxic chemical that contaminates the environment, leading to potential reproductive issues in living organisms.
  • Researchers used two-dimensional fluorescence difference gel electrophoresis to analyze the impact of TCDD on porcine granulosa cells, identifying 75 protein spots that changed in response to the chemical treatment.
  • The study found that cytoskeletal proteins were the most affected, suggesting that TCDD may disrupt ovarian follicle function by altering the cytoskeleton and stress response proteins, providing insights for future research.

Aryl hydrocarbon receptor (AhR) is a ligand-activated transcription factor best known for mediating xenobiotic-induced toxicity. AhR requires aryl hydrocarbon receptor nuclear translocator (ARNT) to form an active transcription complex and promote the activation of genes which have dioxin responsive element in their regulatory regions. The present study was performed to determine the complete cDNA sequences of porcine AhR and ARNT genes and their chromosomal localization.

Article Synopsis
  • Polychlorinated dibenzo-p-dioxins (PCDDs) are harmful by-products of industrial activities that accumulate in living tissues and can have negative effects on health.
  • Dioxins are metabolized by the CYP1A1 enzyme, and their toxicity varies based on the number and position of chlorine atoms in their structure.
  • This study found that TCDD, a specific dioxin, has a strong affinity for CYP1A1 but also significantly impacts the enzyme's function, explaining its long half-life and high toxicity in organisms.
