Genome-wide association studies (GWAS) provide a powerful means to identify loci and genes contributing to disease, but in many cases the related cell types/states through which genes confer disease risk remain unknown. Deciphering such relationships is important for identifying pathogenic processes and developing therapeutics. Here, we introduce sc-linker, a framework for integrating single-cell RNA-seq (scRNA-seq), epigenomic maps and GWAS summary statistics to infer the underlying cell types and processes by which genetic variants influence disease.

Purpose: Proton beam therapy has been found to have enhanced biological effectiveness in targets that contain the boron isotope B, with the alpha particles resulting from the p + B → 3α reaction being hypothesized as the mechanism; in this study, we aimed to elucidate the causes of the enhanced biological effectiveness of proton-boron fusion therapy by performing a detailed Monte Carlo study of the p + B → 3α reaction in a phantom geometry.

Methods: We utilized the Geant4 toolkit to create Monte Carlo particle physics simulations. These simulations consisted of a proton beam with a range 30 mm, creating a Spread-Out Bragg Peak with a modulation width of 10 mm, directed into a water phantom containing a region of boron material.

Background: WHO recently approved a partially effective vaccine that reduces clinical malaria in children, but increased vaccine activity is required to pursue malaria elimination. A phase 1 clinical trial was done in Mali, west Africa, to assess the safety, immunogenicity, and protective efficacy of a three-dose regimen of Plasmodium falciparum sporozoite (PfSPZ) Vaccine (a metabolically active, non-replicating, whole malaria sporozoite vaccine) against homologous controlled human malaria infection (CHMI) and natural P falciparum infection.

Methods: We recruited healthy non-pregnant adults aged 18-50 years in Donéguébougou, Mali, and surrounding villages (Banambani, Toubana, Torodo, Sirababougou, Zorokoro) for an open-label, dose-escalation pilot study and, thereafter, a randomised, double-blind, placebo-controlled main trial.

Purpose: The main purpose of this work was to generate and validate the dosimetric accuracy of proton beams of dimensions that are appropriate for in vivo small animal and in vitro ultrahigh dose rate (FLASH) radiotherapy experiments using a synchrotron-based treatment delivery system. The nutrient-sensor O-GlcNAc transferase (Ogt), the sole enzyme that adds an O-GlcNAc-modification onto proteins, plays a critical role for pancreatic β-cell survival and insulin secretion. We hypothesized that β-cell Ogt overexpression would confer protection from β-cell failure in response to metabolic stressors, such as high-fat diet (HFD) and streptozocin (STZ). Here, we generated a β-cell-specific Ogt in overexpressing (βOgtOE) mice, where a significant increase in Ogt protein level and O-GlcNAc-modification of proteins were observed in islets under a normal chow diet.

An off-axis digital holographic imaging polarimeter was developed to estimate the Jones matrices of an object. The Jones vector image of the electric field returned from the object is determined from a single holographic recording using the interference between the dual, nearly orthogonal, reference beams. The technique compensates for phase variations in the optical beam paths between the recorded holograms and relaxes the need to generate orthogonal illumination polarization states.

Microbial keratitis is devastating corneal morbidity with a variable spectrum of clinical manifestations depending on the infective etiology.
