Article Synopsis
  • Thyroid cancer is the most common type of endocrine cancer and generally has a good outlook, but some cases, particularly follicular-origin cancers, can become aggressive and resistant to treatment.
  • A significant number of these cancers show hyper-activation of the MAPK signaling pathway due to specific mutations, which has led to the testing of targeted therapies, such as BRAF and MEK inhibitors.
  • Current strategies include using these inhibitors to try and redifferentiate refractory tumors, potentially allowing for subsequent treatment with radioactive iodine, but resistance to treatment remains a challenge.

Obesity is a pathophysiological condition where excess free fatty acids (FFA) target and promote the dysfunctioning of insulin sensitive tissues and of pancreatic β cells. This leads to the dysregulation of glucose homeostasis, which culminates in the onset of type 2 diabetes (T2D). FFA, which accumulate in these tissues, are metabolized as lipid derivatives such as ceramide, and the ectopic accumulation of the latter has been shown to lead to lipotoxicity.
