A 32-year-old female with amenorrhea-galactorrhea due to primary hypothyroidism was treated with thyroid hormones, and serum levels of thyrotropin (TSH), prolactin (PRL), triiodothyronine (T3), thyroxin (T4), and T3 resin sponge uptake (RT3U) were measured throughout the course of treatment. The elevated serum levels of TSH and PRL fell into the normal range following T3 treatment. Subsequently, the menstrual cycle was restored within 1 month, and galactorrhea completely ceased and conception was achieved within 3 months.
View Article and Find Full Text PDFA simple device for mixing water and organic solvents can be easily constructed to dehydrate tissue for electron microscopy. The device utilizes hydrostatic leveling between a reservoir and a mixing chamber to produce a continuously increasing concentration of dehydrating solvent. The rate of dehydration may be regulated by geometry and/or outflow rate.

The liver of rats fasted for 48 h is perfused with an erythrocyte-free medium. Ultrastructural analysis shows integrity of all subcellular organelles with the exception of minor alterations in the rough endoplasmic reticulum.

Four patients with Graves' disease whose hyperthyroidism was in remission following antithyroid therapy were studied without any treatment during and after pregnancy. In the 8-9th month of pregnancy, they were in a euthyroid state with serum levels of thyroxine (T4) of 18.9, 11.

Seven of these patients were euthyroid and the other 3 had hypothyroidism. The patients in the euthyroid group received 50 mug of T3 daily for 7 days and 75 mug q.

A linear relationship between the volume of eluate and the TSH value and good recoveries of endogenous TSH (104%) and added TSH (89%) were obtained, indicating that TSH in dried blood was extracted well by overnight elution and determined accurately by radioimmunoassay. The TSH content in the central portion of a spot was less (71%) than that in the peripheral portion.

Rat livers were perfusion fixed with buffered 2 per cent paraformaldehyde or a mixture of 1.5 per cent paraformaldehyde and 1 per cent glutaraldehyde and processed as follows.
View Article and Find Full Text PDFLamellar bodies isolated from rat lung homogenates were prepared for electron microscopy by two methods: OsO4 fixation followed by dehydration either with acetone or ethanol. Electron microscopy revealed that the acetone-dehydrated fraction contained many well preserved lamellar bodies with intact outer membrane, and the high retention of a material of low electron density between the fine and closely spaced concentric lamellae. This appearance was contrasted by the increased loss of this interlamellar material and coarse and irregularly spaced lamellae in the ethanol-dehydrated fraction.

The urinary 17-hydroxycorticosteroids excretion was persistently reduced and was increased by repeated ACTH injections. The low basal metabolic rate, reduced serum thyroid hormones and elevated serum thyrotrophic hormone were consistent with primary hypothyroidism.
