We present flow matching for reaction coordinates (FMRC), a novel deep learning algorithm designed to identify optimal reaction coordinates (RC) in biomolecular reversible dynamics. FMRC is based on the mathematical principles of lumpability and decomposability, which we reformulate into a conditional probability framework for efficient data-driven optimization using deep generative models. While FMRC does not explicitly learn the well-established transfer operator or its eigenfunctions, it can effectively encode the dynamics of leading eigenfunctions of the system transfer operator into its low-dimensional RC space.

Unlabelled: Cytotoxins (CTXs), proteins found in cobra venom, selectively inhibit tumor cell proliferation. Herein, we selected CTX-XII because of its potent antitumor activity to investigate the effect of solution pH on its response. MTT assay results showed significantly higher inhibition rates for CTX-XII at pH 5.

Biomolecular simulations often suffer from the "time scale problem", hindering the study of rare events occurring over extended time scales. Enhanced sampling techniques aim to alleviate this issue by accelerating conformational transitions, yet they typically necessitate well-defined collective variables (CVs), posing a significant challenge. Machine learning offers promising solutions but typically requires rich training data encompassing the entire free energy surface (FES).

  • The study explores how incorporating patient-reported outcomes (PROs) can enhance the prediction of hospitalization and mortality risks in patients with heart failure (HF).
  • The research involved 1165 patients with heart failure with reduced ejection fraction (HFrEF) and 456 with preserved ejection fraction (HFpEF), utilizing advanced statistical methods to analyze risk over time.
  • Findings indicated that models including PROs significantly improved risk prediction, demonstrating their value alongside traditional clinical assessments in managing outpatient heart failure.

Many compartments are prone to pose safety hazards such as loose fasteners or object intrusion due to their confined space, making manual inspection challenging. To address the challenges of complex inspection environments, diverse target categories, and variable scales in confined compartments, this paper proposes a novel GMS-YOLO network, based on the improved YOLOv8 framework. In addition to the lightweight design, this network accurately detects targets by leveraging more precise high-level and low-level feature representations obtained from GhostHGNetv2, which enhances feature-extraction capabilities.

Developing highly effective catalysts for ammonia (NH) synthesis is a challenging task. Even the current, prevalent iron-derived catalysts used for industrial NH synthesis require harsh reaction conditions and involve massive energy consumption. Here we show that anchoring buckminsterfullerene (C) onto non-iron transition metals yields cluster-matrix co-catalysts that are highly efficient for NH synthesis.

Recently discovered epigenetic modification lysine lactylation contributes to tumor development and progression in several types of cancer. In addition to the tumor-intrinsic effects, histone lactylation may mediate tumor microenvironment remodeling and immune evasion. In this study, we observed elevated pan-lysine lactylation and histone H3 lysine 18 lactylation (H3K18la) levels in non-small cell lung cancer (NSCLC) tissues, which was positively correlated with poor patient prognosis.

Background: MiR-486-5p has been identified as a crucial regulator of the PI3K/AKT signalling pathway, which plays a significant role in skeletal muscle development. Its host gene, sANK1, is also essential for skeletal muscle development. However, the understanding of porcine miR-486-5p and sANK1 has been limited.

  • The study aimed to evaluate the effectiveness and tolerability of direct-acting antiviral (DAA) treatments for hepatitis C virus (HCV) across different genotypes (GTs) in a global, real-world context, focusing particularly on GT3 and GT6.
  • Researchers analyzed data from 15,849 chronic hepatitis C patients across Asia, North America, and Europe over a seven-year period, noting demographic factors such as age, sex, and prior treatment history.
  • Results showed a high sustained virological response (SVR12) rate of 96.9% overall, with variances by genotype, highlighting that independent factors like advanced age, cirrhosis, and previous treatment failures affected treatment outcomes, while being

With the rapid development of intensive animal husbandry in the livestock industry, large quantities of manure waste containing phytate phosphorus are being generated. Phytase can effectively solve the problem of high phosphorus pollution in the feces of monogastric animals. Enviropig, which produces phytase in the salivary glands and secretes the enzyme in the saliva, were first generated in 1999.

Ammonia (NH) is closely related to the fields of food and energy that humans depend on. The exploitation of advanced catalysts for NH synthesis has been a research hotspot for more than one hundred years. Previous studies have shown that the Ru B sites (step sites on the Ru (0001) surface uniquely arranged with five Ru atoms) and Fe C sites (iron atoms with seven nearest neighbors) over nanoparticle catalysts are highly reactive for N-to-NH conversion.

Background: Gastrointestinal (GI) function is critical for patients in intensive care units (ICUs). Whether and how much critically ill patients without GI primary diseases benefit from abdominal physical examinations remains unknown. No evidence from big data supports its possible additive value in outcome prediction.

Homotypic membrane fusion of the endoplasmic reticulum (ER) is mediated by dynamin-like GTPase atlastin (ATL). This fundamental process relies on GTP-dependent domain rearrangements in the N-terminal region of ATL (ATL), including the GTPase domain and three-helix bundle (3HB). However, its conformational dynamics during the GTPase cycle remain elusive.

Human cancer cell lines have long served as tools for cancer research and drug discovery, but the presence and the source of intra-cell-line heterogeneity remain elusive. Here, we perform single-cell RNA-sequencing and ATAC-sequencing on 42 and 39 human cell lines, respectively, to illustrate both transcriptomic and epigenetic heterogeneity within individual cell lines. Our data reveal that transcriptomic heterogeneity is frequently observed in cancer cell lines of different tissue origins, often driven by multiple common transcriptional programs.
