Article Synopsis
  • The research addresses challenges in studying ESR2 proteins due to limited detection systems, and introduces a new monoclonal antibody (PPZ0506) that works for both human and rodent ESR2 proteins.* -
  • This study builds on previous work by optimizing immunohistochemical detection methods specifically for mouse ESR2 proteins in paraffin-embedded ovary sections, focusing on critical factors such as heat-induced antigen retrieval and antibody dilution.* -
  • Findings reveal a more localized distribution of mouse ESR2 proteins compared to previous assumptions and highlight interspecies differences when comparing to human and rat ESR2 expression, which could clarify conflicting evidence in the field.*

Although there are few well-validated antibodies against ESR2 proteins, a recent validation assessment identified a specific monoclonal antibody against human ESR2 proteins (PPZ0506). Furthermore, our previous study confirmed its cross-reactivity and specificity against rodent ESR2 proteins, enabling the determination of true ESR2 distribution profiles in rodents. Therefore, we aimed to determine optimal conditions for ESR2 detection by PPZ0506 immunostaining and analyze ESR2 distribution in rats.
