Monoclonal autoantibodies obtained using the native porcine thyroglobulin (Tg) molecule for immunization were shown to react against a defined epitope of the Tg molecule. Its biochemical characterization was undertaken, using trypsin treatment of Tg, followed by gel filtration, affinity chromatography, sodium dodecyl sulfate-polyacrylamide gel electrophoresis and high pressure liquid chromatography analysis. Immunoreactivity tested by immunoblotting with appropriate monoclonal anti-Tg antibody allowed the detection of one peptide fragment of 5-10 kDa, after limited proteolysis with trypsin.

Cultures of synovial cells from normal CBA mice were established after collagenase treatment of synovial tissue collected from the knee joint. Morphological studies using light and electron microscopy have shown that confluent monolayers are composed of 90% triangular or stellate dendritic cells with numerous microvilli and 5% secreting cells containing many dense granules. Less than 5% contaminating cells, such as fibroblasts or macrophages, are present.

Forskolin, a diterpen stimulating the adenylcyclase induces in vitro an accumulation of cAMP in thyroid glands explanted from 17 day-old rat fetuses. Thyroid glands from 15 day-old fetuses exposed to forskolin exhibit, within 24 hr, an active folliculogenesis and 125I fixation in follicular cavities. The results indicate that cAMP probably activates the onset of both morphological and physiological maturation in the fetal rat thyroid.

By using the hybridoma technology, a monoclonal human thyroid cell line was obtained by fusing fresh normal human thyroid cells with a human lymphoblastoid cell line. The resultant cell line, called GEJ, has been selected for its expression of thyrotropin (TSH) receptors and has morphological and functional characteristics of normal human thyroid cells. In the presence or absence of human TSH, the GEJ cell line accumulates iodide, produces thyroid hormones, expresses thyroid membrane antigens, and binds approximately equal to 600 molecules of TSH per cell.

Thyroid glands from 15 day-old rat foetuses were incubated in Eagle's medium containing Na 125I and supplemented, or not, with TSH for 4 or 24 hours. Electron microscopic radioautographic study shows silver grains mainly in follicular cavities only in the thyroids submitted to TSH during 24 hr. A functional differentiation must therefore take place in thyroid cells under TSH stimulation.

Incubation in a culture medium with and without TSH of 16 day-old foetal thyroid glands induces hypertrophy of the Golgi apparatus which may be correlated with a considerable increase in the number of secretory vesicles. A stereological study performed during the first 6 hr of incubation showed that: vesicle secretion was biphasic; vesicle secretion was heterogeneous with two different populations of vesicles; When TSH (20 mU and 80 mU) was added to the medium, the volume density of the follicular lumina increased; at least during the first 6 hr TSH seemed to be necessary to the formation of follicular lumina.

The authors report one case of sinus histiocytosis with massive lymphadenopathy which occurred in a 70 years old woman. They remind the main features of this pseudolymphomatous benign disorder with three key microscopic findings : massive sinus histiocytosis, lymphophagocytosis, proliferation of mature plasma cells. When cultured in polystyrene dishes subjected to previous treatment and supplied with a serum-containing medium, hog thyroid cells form monolayers displaying dome-like arrangements after three to four days. Cells involved in formation of "domes" are morphologically polarized; the apical microvilli of these cells point toward the culture medium. When the tissue is cultured in untreated polystyrene dishes, thyroid cells remain in suspension; their aggregates swell progressively and form hollow spheres encompassed by a single layer of cells.

Thirty-two thyroid tumors, 9 benign, 23 malignant, and 12 samples of normal thyroid tissue were examined by light and electron microscopy. Thyroglobulin content was also measured in the tissues and, in a limited number of cases, enzymatic activities were determined, such as thyroid peroxidase-iodinase, acid protease, and deiodinase. The presence of significant amounts of 19S, 27S and 12S thyroglobulin was well correlated with the ability of the tumors to accumulate radioiodine.

A human thyroid tumor is described which, on light microscopy, exhibited the features of medullary carcinoma of the thyroid (MCT). The cells were arranged in solid lobules or trabeculae, and the stroma was abundant and gave positive reaction for amyloid as assessed by congo red, crystal violet and thioflavin T stains. However, there was microfollicular differentiation in certain portions of the tumor even where the tumor was invasively growing.

Three thyroid carcinomas composed in part or entirely of clear cells were studied by light and electron microscopy, and thyroglobulin content was determined by biochemical methods. Clear cells have been found in follicular and papillary thyroid carcinomas and in undifferentiated carcinomas.
