Impaired epithelial barrier function is a hallmark of inflammatory bowel diseases, such as colitis, contributing to diarrhoea and perpetuating inflammation. We show that the zinc sensing receptor, ZnR/GPR39, triggers intracellular Ca(2+) signalling in colonocytes thereby inducing occludin expression. Moreover, ZnR/GPR39 is essential for epithelial barrier recovery in the dextran sodium sulfate (DSS) ulcerative colitis model.

Article Synopsis
  • Utilizing endogenous molecules like α1-antitrypsin (AAT) is generally more effective as a therapy than synthetic alternatives, particularly due to its anti-inflammatory and wound-healing properties.
  • AAT has been used for decades to treat a rare genetic deficiency and shows promise in reducing tissue damage from conditions such as ischemic heart disease and stroke.
  • Current clinical trials are investigating AAT for treating type 1 diabetes, with early results indicating it may enhance insulin production without negative side effects for some patients.
