Antidrug antibodies (ADAs) against biologics present a major challenge for sustained biotherapy, including enzyme replacement therapies and adeno-associated virus (AAV) gene therapies. These antibodies arise from undesirable immune responses, leading to altered pharmacokinetics, reduced efficacy, and adverse reactions. In this study, we introduced a rationally designed lipid-rapamycin (Rapa)-based nanovaccine to restore immune tolerance to biologics and overcome drug resistance.

The confining effect is essential to regulate the activity and stability of single-atom catalysts (SACs), but the universal fabrication of confined SACs is still a great challenge. Here, various lattice-confined Pt SACs supported by different carriers are constructed by a universal co-reduction approach. Notably, Pt single atoms confined in the lattice of Ni(OH) (Pt/Ni(OH)) with a high electron-deficient state exhibit excellent activity for basic hydrogen evolution reaction (HER).

  • Transmissible gastroenteritis virus (TGEV) is a harmful coronavirus that can lead to deadly intestinal diseases in young pigs, particularly those under two weeks old, with a lethal outcome rate of 100%.
  • The research utilized monocyte-derived porcine dendritic cells (DCs) to examine how TGEV interacts with these important immune cells, revealing that TGEV can replicate in these cells while UV-inactivated TGEV cannot.
  • Findings showed that TGEV infection significantly raised the expression of SLA-DR, a crucial molecule for immune response, and that the ORF3b protein of TGEV enhances this expression at a transcriptional level, shedding light on the virus's pathogenic mechanisms.

As the most abundant post-transcriptional modification in eukaryotes, N-methyladenosine (mA) plays a crucial role in cancer cell proliferation, invasion and chemoresistance. However, its specific effects on chemosensitivity to oxaliplatin-based regimens and the impact of these drugs on mA methylation levels in colorectal cancer (CRC) remain largely unexplored. In this study, we demonstrated that the mA methyltransferase Wilms tumor 1-associating protein (WTAP) weakens oxaliplatin chemosensitivity in HCT116 and DLD1 cells.

To reduce the cost of docosahexaenoic acid (DHA) production from Schizochytrium sp., the waste Pichia pastoris was successfully used as an alternative nitrogen source to achieve high-density cultivation during the cell growth phase. However, due to the high oxygen consumption feature when implementing high-density cultivation, the control of both the nitrogen source and dissolved oxygen concentration (DO) at each sufficient level was impossible; thus, two realistic control strategies, including "DO sufficiency-nitrogen limitation" and "DO limitation-nitrogen sufficiency", were proposed.

  • Cancer cells rely on metabolic adaptations to survive oxidative stress, and this study focuses on how specific enzymes, particularly MTHFD2, protect gastric cancer cells from high levels of reactive oxygen species (ROS).
  • The researchers utilized various biological methods and models, including patient-derived xenografts, to show that overexpression of MTHFD2 is linked to worse survival outcomes and that its knockdown leads to increased ROS and cell death under hypoxic conditions.
  • Findings suggest that targeting MTHFD2 could be a promising therapeutic approach for treating gastric cancer by disrupting its redox balance and reducing tumor growth.

Porcine deltacoronavirus (PDCoV) and porcine sapelovirus (PSV) are two viruses that can cause diarrhoea in pigs and bring great economic loss to the pig industry. In this research, a duplex real-time quantitative polymerase chain reaction (qPCR) assay based on SYBR Green I was developed to simultaneously detect PDCoV and PSV. No specific melting peaks were found in other porcine diarrhoea-associated viruses, indicating that the method developed in this study had good specificity.

Media exposure to health communication contents related to the COVID-19 pandemic alone is inadequate to measure the influence of media on individuals in adopting precautionary behaviors against COVID-19, such as vaccine uptake. Certain individuals may pay attention to and be influenced by communication content. However, literature has suggested other instrumental determinants in developing and adopting health precautionary measures, such as culture or religion, especially regarding vaccination.

Cellular omics such as single-cell genomics, proteomics, and microbiomics allow the characterization of tissue and microbial community composition, which can be compared between conditions to identify biological drivers. This strategy has been critical to revealing markers of disease progression, such as cancer and pathogen infection. A dedicated statistical method for differential variability analysis is lacking for cellular omics data, and existing methods for differential composition analysis do not model some compositional data properties, suggesting there is room to improve model performance.
