Protein liquid-liquid phase separation (LLPS), a pivotal phenomenon intricately linked to cellular processes, is regulated by various other proteins. However, there is still a lack of high-throughput methods for screening protein regulators of LLPS in target proteins. Here, we developed a CRISPR/Cas9-based screening method to identify protein phase separation regulators by integrating bimolecular fluorescence complementation (BiFC) and fluorescence-activated cell sorting (FACS).

Transcriptome-wide alternative polyadenylation (APA) is involved in both innate and adaptive immune responses of immune cells. Downregulation of the CPSF6 protein, one of the 3' end-processing factors, mediates APA in macrophages with responses to virus infection and plays an important role in its anti-virus effect. However, the signaling pathway and molecular mechanism underlying the downregulation of the CPSF6 protein remain elusive.

This study investigates the number line estimation (NLE) strategies utilized by children aged 4-7 across both bounded and unbounded NLE tasks. Drawing on prior research, it hypothesized that younger children would predominantly employ benchmark-based strategies, with endpoints as reference points, in bounded tasks, while older children would utilize a wider range of reference points including midpoints and quartiles. For unbounded tasks, it was anticipated that both younger and older children would adopt the scalloped strategy.

Article Synopsis
  • Vaccine efficacy is influenced by both antigens and adjuvants, which enhance immune responses; however, current adjuvants don't effectively work with live attenuated viruses.
  • Researchers developed a recombinant virus (WH2020-ΔTK/gI/gE-G-CSF) that expresses feline G-CSF, demonstrating similar growth patterns to its predecessor and improving immune responses.
  • Kittens vaccinated with the G-CSF-expressing virus produced more neutralizing antibodies and neutrophils, leading to milder clinical symptoms after FHV-1 infection, highlighting G-CSF's potential as an immune booster for live vaccines.

Neoadjuvant immunochemotherapy (NAIC) achieves superior clinical benefits over neoadjuvant chemotherapy (NAC) in multiple types of human cancers, including gastric adenocarcinoma (GAC). However, it is poorly understood how the malignant epithelial cells and tumor immune microenvironment (TIME) might respond distinctly to NAIC and NAC that underlies therapeutic efficacy. Here treatment-naive and paired tumor tissues from multiple centers were subjected to pathological, immunological, and transcriptomic analysis.

Article Synopsis
  • The study examines the impact of tumor budding (TB) as a prognosis factor in stage II colorectal cancer (CRC) patients who underwent surgery, analyzing how its features relate to outcomes in postoperative chemotherapy.
  • Data from 336 patients, categorized into three TB grades (BD1, BD2, BD3), reveal that higher TB grades correlate with lower rates of recurrence-free survival (RFS) and overall survival (OS), especially highlighting BD3 patients' poor outcomes.
  • While adjuvant chemotherapy significantly benefits patients with lower TB grades (BD1/2), it does not improve survival for BD3 patients, suggesting BT's role as a crucial predictor of treatment efficacy and prognosis.
