Trametinib, an inhibitor of mitogen-activated extracellular signal-regulated kinases 1/2 (MEK1/2), is used to treat BRAF melanoma and non-small-cell lung cancer (NSCLC). Mutant Kirsten rat sarcoma viral oncogene homolog (KRAS) promotes glutamine utilization, therefore, in the present study we investigated the anti-cancer effects of trametinib in combination with V-9302, a glutamine transporter inhibitor, in NSCLC with KRAS mutations. Trametinib in combination with V-9302 exhibited a potent synergistic antitumor effect, inducing cell cycle arrest and pyroptosis.

Although the glycosylation of viral proteins plays a critical role in the process of viral invasion into host cells, studies on the glycosylation of monkeypox virus (MPXV) structural proteins have not yet been reported. To investigate the importance of MPXV protein glycosylation, poly Ser-Arg (poly SR) materials capable of simultaneously enriching both -glycopeptides and -glycopeptides were synthesized by surface-initiated reversible addition-fragmentation chain transfer (SI-RAFT) polymerization. The poly SR materials were evaluated using the digest mixture of standard proteins containing bovine fetuin and bovine serum albumin, and the digest of complex biological samples including bovine sperm tail lysate, mouse sperm tail lysate, mouse brain lysate, and human serum.

Senescence of nondividing neurons remains an immature concept, with especially the regulatory molecular mechanisms of senescence-like phenotypes and the role of proteins associated with neurodegenerative diseases in triggering neuronal senescence remaining poorly explored. In this study, we reveal that the nucleolar polyglutamine binding protein 3 (PQBP3; also termed NOL7), which has been linked to polyQ neurodegenerative diseases, regulates senescence as a gatekeeper of cytoplasmic DNA leakage. PQBP3 directly binds PSME3 (proteasome activator complex subunit 3), a subunit of the 11S proteasome regulator complex, decreasing PSME3 interaction with Lamin B1 and thereby preventing Lamin B1 degradation and senescence.
