Article Synopsis
  • Alzheimer's disease (AD) is a severe neurodegenerative disorder common in older adults, and cholinesterase inhibitors (ChEI) are effective palliative treatments for it.
  • Researchers synthesized new thiosemicarbazone derivatives to target cholinesterase enzymes and evaluated their biological activity, particularly against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE).
  • Compounds 18 and 19 showed strong inhibition against BChE, while compounds 25, 26, and 28 demonstrated superior antioxidant capabilities, especially in cupric reducing assays; compound 19 may compete effectively with the drug galantamine.

The first synthesis of all five possible monomethylated [4]dendralenes has been achieved via two distinct synthetic strategies. The Diels-Alder chemistry of these new dendralenes (as multidienes) with an electron poor dienophile, N-methylmaleimide (NMM), has been studied. Thus, simply upon mixing the dendralene and an excess of dienophile at ambient temperature in a common solvent, sequences of cycloadditions result in the rapid generation of complex multicyclic products.

Article Synopsis
  • The text discusses the dendralenes, a group of acyclic hydrocarbons known for their ability to create complex structures, which has garnered synthetic interest.
  • It highlights the authors' successful synthesis of the "higher" dendralenes, specifically from [9]dendralene to [12]dendralene, as previously, only lower members were documented.
  • The research also explores the unique spectroscopic properties and chemical behaviors of these dendralenes, noting a trend of diminishing alternation in their reactivity and properties linked to their structural conformations.
