Introduction: The Göttingen Minipig (GM) is used as large animal model in articular cartilage research. The aim of the study was to introduce osteoarthritis (OA) in the GM by resecting the anterior cruciate ligament (ACLR) according to Pond and Nuki, verified by histological and magnetic resonance imaging (MRI) scoring as well as analysis of gene and protein expression.
Materials And Methods: The eight included skeletally mature female GM were assessed after ACLR in the left and a sham operation in the right knee, which served as control.
Background: The clinical application of bone morphogenetic proteins such as BMP-2 and GDF-5 (growth and differentiation factor-5) may improve the outcome of bone defect repair. In addition to the osteoinductivity of BMPs, their angiogenic potential is important as an adequate blood supply is a prerequisite for bone-healing. We used a rabbit long-bone defect model to investigate whether angiogenicity and osteogenicity were correlated features of a BMP molecule by comparing the induction of blood vessel and bone formation by BMP-2, GDF-5, and a previously created swap mutant GDF-5V453/V456 (BB-1) with elevated BMP receptor-IA binding.

Peptides generated upon degradation of mitochondrial proteins by various ATP-dependent proteases are continuously released from mitochondria raising the intriguing possibility of a role of these peptides in interorganellar communication. Here, we have determined genome-wide transcript profiles of mutant yeast cells defective in mitochondrial peptide export. Deletion of YME1, coding for the i-AAA protease in the inner membrane, abolished peptide generation in the intermembrane space and led to the induction of nuclear genes with functions in mitochondrial gene expression and the biogenesis of the respiratory chain.
