Imipramine-induced ex copula ejaculates (11) and fractionated in copula ejaculates were collected from each of 5 pony stallions for freezing in 5-ml straws (6), using a modified Kenney glucose skim-milk extender (2). Initial post-thaw total and progressive motilities and daily post-thaw total and progressive motilities, as well as the number of days to reach 0 progressively motile spermatozoa, were also similar for the 2 methods of collection. The percentage of morphologically normal spermatozoa both before freezing and after thawing were also similar for in copula and ex copula ejaculates.

Matrilysin (PUMP, MMP-7) is a member of the metalloprotease gene family, whose constituents are responsible for the remodeling of extracellular matrix. The matrilysin protein is a 28-kDa zymogen possessing catalytic activities against a broad range of extracellular matrix substrates including proteoglycans, gelatin, fibronectin, laminin, and elastin. To gain insights into the biological expression of matrilysin in human cell types, we generated a monospecific, polyclonal antibody against a 16-amino acid sequence derived from its catalytic domain, a region which lacked significant homology with other matrix metalloenzymes.

A Thoroughbred stallion with erectile dysfunction following paraphimosis was managed to allow consistent ejaculation. Ejaculation was elicited in this stallion by applying manual stimulation to the base of the penis. Two mature breeding stallions were evaluated because of specific ejaculatory dysfunction, and each was found to have aortic-iliac thrombosis occluding 60 to 70% of the aortic lumen. In each case, the stallion had strong libido, normal mounting, and vigorous initial thrusting. With continued exertion, thrusting became weak and dismount was awkward.

Emission and bladder neck closure are primarily alpha-adrenergically mediated thoracolumbar sympathetic reflex events with supraspinal modulation. A 9-year-old atactic breeding stallion was trained to ejaculate, with only manual stimulation, while standing on the ground. Ejaculates obtained yielded fertile semen with morphologic and motility characteristics within the range for normal stallions. This method extended the breeding life of a stallion unable to mount a live or dummy mare or to ejaculate into an artificial vagina while standing on the ground.

The expression of the metalloproteinase stromelysin correlates with the progression of chemically induced squamous cell carcinomas. We demonstrate that the expression of activated stromelysin in papilloma-derived cells enhances in vitro cell invasion. The expression of members of the family of matrix-degrading metalloproteinases (MMPs) is believed to contribute to the complex process of invasion and metastasis. In this study, specific cDNA probes for three members of the stromelysin subfamily of MMPs--stromelysin (MMP-3), stromelysin-2 (MMP-10), and pump-1 (MMP-7)--were used to examine the expression of these three different MMPs in human gastric and colonic carcinomas and in adjacent normal mucosa. The mating behaviour of 2 jacks, each with 21 non-pregnant jennies, was studied when the jennies were in natural oestrus and simultaneously induced oestrus. The main observations were: efficient pasture breeding at natural and induced oestrus, a territorial sociosexual structure, prolonged pre-copulatory interaction, gradual increase of mating activity up to 2 days before ovulation, a copulatory sequence similar to that of horses, vocalization of the jack as a conspicuous behaviour initiating pre-copulatory interaction, frequent heterotypical behaviour of jennies and active involvement of jennies in initiating pre-copulatory interaction.

There are several characteristics of stromelysin that suggest that expression of this enzyme may play an important role in tumor invasion and metastasis; the stromelysin gene is expressed in response to stimulation by oncogenes and tumor promoters, and the protein product of this gene is a metalloproteinase capable of degrading multiple components of the extracellular matrix. Experimental evidence to support this hypothesis has been derived from several animal model systems, in which a positive correlation has been observed between stromelysin expression and tumor progression and metastasis. In addition, in vivo experiments in which the levels of TIMP, the tissue inhibitor of metalloproteinases, were altered also strongly suggest a causal role for metalloproteinases in tumor metastases.

Stromelysin (transin) is a secreted metalloprotease that is transcriptionally induced by a variety of growth factors and oncogenes. We examined the necessity of specific secondary (protein kinase C) and tertiary (c-fos and c-jun protein products) messengers in the transactivation of stromelysin gene expression by epidermal growth factor (EGF). Rat-1 fibroblasts exposed to antisense c-fos DNA or RNA demonstrated that c-fos expression was necessary for complete EGF induction of stromelysin expression.

We have raised an mAb to a previously undescribed 135-kD externally disposed integral membrane protein that is enriched in the intercellular junctional domain of cultured human umbilical vein endothelial cells. This protein localizes at the appositional surfaces of cells as they become confluent and is stably expressed in the junctional zones of confluent monolayers. This protein is expressed in situ on continuous endothelia of all blood vessels in all human tissues examined.

Twelve pony geldings with (n = 6) and without (n = 6) testosterone replacement (200 micrograms/kg testosterone propionate in oil, SC every 48 hours) received either gonadotropin releasing hormone (GnRH; 25 micrograms SC every 3 hours) or control treatment. Sexual behavior was recorded during 4-minute exposure to an estrous mare, 3 times weekly for 2 weeks before treatment, 3 weeks during treatment, and 3 weeks after treatment had been discontinued. The group receiving testosterone and GnRH (n = 3) exhibited significantly greater flehmen response frequency and attention duration and significantly lower vocalization frequency and erection duration than the group receiving only testosterone (n = 3).

Series of experiments and clinical trials were conducted to evaluate the effects of psychoneurotropic agents on sexual behaviour of stallions. The benzodiazepine derivative, diazepam (Valium), effectively reversed experimentally suppressed precopulatory arousal and response. Diazepam treatment also blocked the negative effect of novel environment on sexual response.
