Article Synopsis
  • Melanoma antigen gene (MAGE)-type antigens are effective targets for cancer immunotherapy, as they appear in cancer cells but not in normal tissues, with the exception of some male germline cells.
  • Researchers focused on the mouse P1A antigen, identifying a specific CD8 T-cell epitope presented by the H-2D molecule in C57BL/6 mice, using adenovirus and modified vaccinia Ankara vaccines to enhance immune responses.
  • The study successfully induced a strong immune response targeting a specific 9-amino acid peptide from the P1A antigen, leading to protection against specific tumors and the identification of T-cell receptors (TCRs) that could be used for adoptive cell therapy.

Life on Earth uses DNA as the central template for self-replication, genetic encoding, and information transfer. However, there are no physical laws precluding life's existence elsewhere in space, and alternative life forms may not need DNA. In the search for exobiology, knowing what to look for as a biosignature remains a challenge - especially if it is not from the obvious list of biologic building blocks.

Cecal volvulus is a rare and life-threatening cause of intestinal obstruction with multiple risk factors including prior abdominal surgery and cecal hypermobility. Although its incidence has been reported after common procedures such as cholecystectomy and appendectomy, it has not been well studied after laparoscopy, especially in gynecological surgeries. If untreated, a cecal volvulus can result in serious complications such as intestinal strangulation, necrosis, or perforation.

Article Synopsis
  • A study investigated the safety of outpatient total thyroidectomy in 183 patients with hyperthyroidism over a seven-year period, focusing on complications and postoperative outcomes.
  • Results showed no instances of thyroid storm, with low rates of complications like vocal nerve damage and hypocalcemia, along with minimal emergency department visits and no readmissions.
  • The findings suggest that outpatient surgery could be a viable option for hyperthyroid patients, highlighting the need for further research to understand factors influencing postoperative risks.

Tumor cell intravasation is essential for metastatic dissemination, but its exact mechanism is incompletely understood. We have previously shown that in breast cancer, the direct and stable association of a tumor cell expressing Mena, a Tie2/VEGF macrophage, and a vascular endothelial cell, creates an intravasation portal, called a "tumor microenvironment of metastasis" (TMEM) doorway, for tumor cell intravasation, leading to dissemination to distant sites. The density of TMEM doorways, also called TMEM doorway score, is a clinically validated prognostic marker of distant metastasis in breast cancer patients.

The physiology and pathophysiology of the pancreas are complex. Diseases of the pancreas, such as pancreatitis and pancreatic adenocarcinoma (PDAC) have high morbidity and mortality. Intravital imaging (IVI) is a powerful technique enabling the high-resolution imaging of tissues in both healthy and diseased states, allowing for real-time observation of cell dynamics.

Delivery of self-amplifying mRNA (SAM) has high potential for infectious disease vaccination due its self-adjuvating and dose-sparing properties. Yet a challenge is the susceptibility of SAM to degradation and the need for SAM to reach the cytosol fully intact to enable self-amplification. Lipid nanoparticles have been successfully deployed at incredible speed for mRNA vaccination, but aspects such as cold storage, manufacturing, efficiency of delivery, and the therapeutic window would benefit from further improvement.

Prostate cancer is a leading cause of cancer death in men. Inflammation and overexpression of inducible nitric oxide synthase (NOS2) have been implicated in prostate carcinogenesis. We aimed to explore the hypothesis that nitric oxide NO exerts pro-tumorigenic effects on prostate cells at physiologically relevant levels contributing to carcinogenesis.

Pancreatitis and pancreatic ductal adenocarcinoma (PDAC) are grave illnesses with high levels of morbidity and mortality. Intravital imaging (IVI) is a powerful technique for visualizing physiological processes in both health and disease. However, the application of IVI to the murine pancreas presents significant challenges, as it is a deep, compliant, visceral organ that is difficult to access, easily damaged and susceptible to motion artefacts.

Article Synopsis
  • Pancreatic ductal adenocarcinoma (PDAC) is difficult to treat due to its ability to evade immune responses, but a new microbiome-based therapy shows promise by introducing a tetanus toxoid protein into tumor cells, reactivating existing T cell responses.
  • In mouse models, this treatment led to the accumulation of the tetanus protein in tumors, recruitment of CD4 T cells, and the production of immune molecules that help kill tumor cells, especially when combined with low doses of gemcitabine (GEM).
  • The combination treatment significantly reduced tumor size (by 80%) and metastases (by 87%), while increasing survival rates in treated mice, suggesting this approach could be a viable alternative for

Cyanuric acid (CYA) is used commercially for maintaining active chlorine to inactivate microbial and viral pathogens in swimming pools and hot tubs. Repeated CYA addition can cause a lack of available chlorine and adequate disinfection. Acceptable CYA levels can potentially be restored via cyanuric acid hydrolases (CAH), enzymes that hydrolyze CYA to biuret under mild conditions.

The degree of metastatic disease varies widely among patients with cancer and affects clinical outcomes. However, the biological and functional differences that drive the extent of metastasis are poorly understood. We analyzed primary tumors and paired metastases using a multifluorescent lineage-labeled mouse model of pancreatic ductal adenocarcinoma (PDAC)-a tumor type in which most patients present with metastases.

Background: The clinical benefit of immune checkpoint blockade (ICB) therapy is often limited by the lack of pre-existing CD8 T cells infiltrating the tumor. In principle, CD8 T-cell infiltration could be promoted by therapeutic vaccination. However, this remains challenging given the paucity of vaccine platforms able to induce the strong cytotoxic CD8 T-cell response required to reject tumors.

Human endogenous retroviruses (HERV), ancient integrations of exogenous viruses, make up 8% of our genome. Long thought of as mere vestigial genetic elements, evidence is now accumulating to suggest a potential functional role in numerous pathologies including neurodegenerative diseases, autoimmune disorders, and multiple cancers. The youngest member of this group of transposable elements is HERV-K (HML-2).
