Molecular refinement of the 1p36 deletion syndrome reveals size diversity and a preponderance of maternally derived deletions.

The deletion of chromosome 1p36 is a newly recognized, relatively common contiguous gene deletion syndrome with a variable phenotype. The clinical features have recently been delineated and molecular analysis indicates that the prevalence of certain phenotypic features appears to correlate with deletion size. Phenotype/genotype comparisons have allowed the assignment of certain clinical features to specific deletion intervals, significantly narrowing the regions within which to search for candidate genes.

Expression of two consecutive genes of a secondary metabolic pathway in transgenic tobacco: molecular diversity influences levels of expression and product accumulation.

We have created a population of transgenic tobacco plants carrying cDNAs encoding two consecutive enzymes from early stages in monoterpenoid alkaloid biosynthesis in Catharanthus roseus. The cDNAs, encoding tryptophan decarboxylase (tdc) and strictosidine synthase (str1) together with a selectable marker gene, were introduced on a single transforming plasmid into tobacco leaves by particle bombardment. Analysis of 150 independent transgenic plants at the DNA and RNA levels demonstrated a range of integration events and steady-state transcript levels for the tdc and str1 transgenes.

Urinary bladder marsupialization for treatment of obstructive urolithiasis in male goats.

Objectives: To describe a surgical procedure for urinary bladder marsupialization and to report the results obtained from its use in the treatment of obstructive urolithiasis in male goats.

Study Design: Retrospective evaluation.

Animals Or Sample Population: Male goats with obstructive urolithiasis.

Evaluation of mild lameness in horses trotting on a treadmill by clinicians and interns or residents and correlation of their assessments with kinematic gait analysis.

Objective: To estimate sensitivity and accuracy of subjective evaluation of mild lameness in horses during treadmill locomotion and to correlate subjective evaluation with kinematic analysis.

Animals: 19 lame and 5 clinically normal horses.

Procedure: Lameness was evaluated by subjective score and kinematic analysis before and after palmar digital nerve block (PDNB).

Implant and provisional crown placement: a one stage protocol.

Most endosseous implant systems are placed using a two-stage surgical procedure. The surgical stages are separated by a period of 4 to 6 months to allow osseointegration. At Stage-I surgery, the implant is fixed in bone and submerged under mucosa, while at Stage-II surgery, the implant is exposed and the transmucosal abutment is connected to the implant.

Bond strengths of conventional and simplified bonding systems.

Purpose: To compare the shear bond strengths of composite to dentin using conventional (three-component) and simplified (two-component) adhesive systems.

Materials And Methods: 100 bovine teeth were mounted in phenolic rings and ground to 600-grit to obtain 90 flat facial dentin surfaces and 10 flat facial enamel surfaces. The dentin specimens were assigned to nine treatment groups of 10 teeth each.

Precision of fit: the Procera AllCeram crown.

Statement Of Problem: Strength, color stability, and precision of fit are requirements for all-ceramic restorations. The Procera AllCeram crown system, composed of a densely sintered high-purity alumina core combined with a low fusing surface porcelain, appears to satisfy most of these requirements. However, evaluation of marginal fit has not been reported.

The myosin ATPase inhibitor 2,3-butanedione-2-monoxime (BDM) inhibits tip growth and cytokinesis in the fission yeast, Schizosaccharomyces pombe.

The growth of fission yeast cultures was reversibly inhibited by exposure to the myosin-ATPase inhibitor 2,3-butanedione-2-monoxime (BDM). Wild-type cells treated with 20 mM BDM for approximately two generation times were smaller than untreated controls and had a septation index approximately twice that seen in the absence of the inhibitor. The organization of actin at the cell poles was somewhat disorganized in the presence of BDM; however, cells formed a cytokinetic actin ring.

Regulation of ciliary beat frequency by both PKA and PKG in bovine airway epithelial cells.

Ciliary beating is required for the maintenance of lung mucociliary transport. We investigated the role of cyclic nucleotide-dependent protein kinases in stimulating ciliary beat frequency (CBF) in bovine bronchial epithelial cells (BBECs). cAMP-dependent protein kinase (PKA) activity and cGMP-dependent protein kinase (PKG) activity were distinguished after DEAE-Sephacel chromatography of BBEC extracts.

The effects of restorative material and location on the detection of simulated recurrent caries. A comparison of dental film, direct digital radiography and tuned aperture computed tomography.

Objectives: To explore the effects of restorative material and lesion location on the detection of recurrent caries using intra-oral film, direct digital radiography and unprocessed and iteratively restored tuned aperture computed tomography (TACT) images.

Methods: An in vitro model with simulated lesions in half the surfaces studied was used. Lesions of varying sizes were created at either the intersection of the facial or lingual wall and the gingival floor or on the gingival floor midway between the facial and lingual walls in the proximal boxes of 24 molar teeth with MOD inlay preparations that had been restored with amalgam, radiopaque composite or radiolucent composite.

Clinical evaluation of two carbamide peroxide tooth-whitening agents.

A blinded study was conducted to evaluate the efficacy and safety of a dentist-prescribed, accelerated carbamide peroxide tooth-whitening system. Fifty-one patients with discolored teeth completed a clinical trial using an overnight bleaching regimen. One group used an experimental bleaching (whitening) regimen with 10% carbamide peroxide bleaching paste, and another group used the Colgate Platinum Professional Overnight Whitening System.

Identification and characterization of small molecule functional antagonists of the CCR1 chemokine receptor.

The CC chemokines macrophage inflammatory protein-1alpha (MIP-1alpha) and RANTES (regulated on activation normal T cell expressed) have been implicated in rheumatoid arthritis and multiple sclerosis. Since their effects are mediated through the CCR1 chemokine receptor, we set up a small molecule CCR1 antagonist program to search for inhibitors. Through high capacity screening we discovered a number of 4-hydroxypiperidine compounds with CCR1 antagonist activity and report their synthesis and in vitro pharmacology here.

Periotest method: implant-supported framework fit evaluation in vivo.

Statement Of Problem: In implant prosthodontics an accurate fit of the framework to the supporting implants is paramount. However, microgaps occur, unknown to the clinician until complications arise that implicate errors in fit. Therefore prosthodontics would welcome a tool or instrument that provides an objective evaluation of the fit at the implant prosthodontic interface.

Pulsed dye laser lithotripsy for treatment of urolithiasis in two geldings.

Transendoscopic pulsed dye laser lithotripsy was effective in the treatment of calcium carbonate urothlithiasis in 2 adult geldings. Perineal urethrotomy provided convenient access for standing transendoscopic lithotripsy and evacuation of a large cystic calculus in one gelding. In the second horse, an obstructive urethral calculus was fragmented and removed by a transurethroscopic approach.

Effect of omentectomy on adhesion formation in horses.

Objective: To determine if omentectomy would decrease the frequency of postoperative intraabdominal adhesions.

Study Design: Retrospective study.

Animals Or Sample Population: 44 horses that had either two ventral median celiotomies or a ventral median celiotomy and a necropsy more than 4 days later; 19 of these horses had their omentum removed at the initial surgery.

Yeast myosin II: a new subclass of unconventional conventional myosins?

Myosin II is the founder member of a large and structurally diverse clan of actin-based motor proteins. The native myosin II molecule is a hexamer consisting of two heavy chains, two essential light chains (ELC), and two regulatory light chains (RLC). For convenience, the myosin IIs are often subdivided into four subclasses: vertebrate skeletal and cardiac muscle myosin II form one subclass, vertebrate smooth muscle and nonmuscle myosin II a second, invertebrate muscle a third, and protozoan myosin II a fourth [Sellers and Goodson, 1995].

Shear bond strengths of one-bottle dentin adhesives using multiple applications.

This study evaluated the effect of multiple applications of the one-bottle adhesives Prime & Bond, One-Step, and Tenure Quik on the shear bond strength of composite to dentin. In addition, the study examined the effects of surface moisture and dual-cure composite on bond strengths of One-Step and Tenure Quik respectively. The adhesive systems were bonded to the occlusal dentin of extracted human teeth.

This study compared the diagnostic efficacy of four imaging modalities for the detection of artificially induced recurrent caries: intraoral film, direct digital bitewing images, tuned apertuce computed tomography (TACT) slices and iteratively restored TACT images using an in vitro model. Twenty-four posterior teeth were prepared for MOD inlay restorations. These were then restored with different restorative materials (amalgam, Herculite and Durafill).
