Biological studies of analogues of glucose iodinated in positions 1, 2, or 3.

Analogues of glucose labeled with 123 iodine in positions 1, 2 or 3 have been synthesized. The aim of this study was to examine their biological behavior in four experimental models in order to assess whether they could be used to evaluate the uptake of glucose with single photon emission computed tomography (SPECT). The results obtained have shown that none of these molecules enters the cell using the glucose transporter.

[Oxidative stress and apoptosis].

Programmed cell death or apoptosis is a process characterized by several morphological, biochemical and molecular events in response to physiological or pathological stimuli, such as gamma radiation. Free radicals being involved in many physiological and pathological processes, the aim of this study is to investigate the literature about the involvement of oxidative pathway during apoptotic process. As reported by several authors, the literature is abundant in this field and show the complexity of the network in which numerous molecules can regulate cell death and proliferation.

Season of birth and Alzheimer's disease: a population-based study in Saguenay-Lac-St-Jean/Québec (IMAGE Project).

The birth distribution of 399 cases of Alzheimer's disease (AD) identified in the region of Saguenay-Lac-St-Jean (Québec) was compared with that of: (a) the population currently living in the area; and (b) the population born during the same period in the same area. AD cases have been recruited since 1986 by the IMAGE Project. Cases and controls were grouped according to the month of birth and according to the day of birth using density estimation.

The gene responsible for a severe form of peripheral neuropathy and agenesis of the corpus callosum maps to chromosome 15q.

Peripheral neuropathy with or without agenesis of the corpus callosum (ACCPN) is a devastating neurodegenerative disorder that is transmitted as an autosomal recessive trait. Genealogical studies in a large number of affected French Canadian individuals suggest that ACCPN results from a single founder mutation. A genomewide search using 120 microsatellite DNA markers in 14 French Canadian families allowed the mapping of the ACCPN gene to a 5-cM region on chromosome 15q13-q15 that is flanked by markers D15S1040 and D15S118.

Intra- and inter-subject variabilities of CGP 33101 after replicate single oral doses of two 200-mg tablets and 400-mg suspension.

Purpose: The purpose of this study was to use a replicate designed trial to assess the overall, intra- and inter-subject variabilities in pharmacokinetic parameters of CGP 33101 after oral administration of tablets relative to that of powder suspended in water, and to determine the relative proportion of the intra-subject variance to the overall variability.

Methods: Sixteen healthy subjects were randomly assigned to four groups to receive tablets and suspension twice in four different treatment sequences. The plasma concentration-time profile of CGP 33101 was characterized in terms of Cmax, Tmax, and AUC.

Experimental models, protocols, and reference values for evaluation of iodinated analogues of glucose.

For an iodinated analogue of glucose to be useful for evaluating glucose uptake using single-photon emission computed tomography (SPECT), it must enter the cell via the same transporter as glucose and accumulate within the cell without being degraded. The biological behavior of the iodinated tracer must therefore be similar to that of 2-deoxy-D(-)[1-14C]-glucose (2-DG). In the present study, four experimental models (biodistribution in mouse, isolated rat heart, human erythrocytes in suspension and cultured neonatal rat cardiomyocytes) have been chosen and protocols have been set up which allow for the examination of small quantities of iodinated analogues of glucose.

Water retention of treated stratum corneum measured by a coupling method: thermal desorption-mass spectrometry.

Synopsis A thermal desorption autosampler coupled to a mass spectrometer has been used to measure the in vitro water retention of human stratum corneum as a function of treatment applied. Samples were treated with ingredients possessing good hygroscopic properties which are used in the formulation of moisturizing creams. After being treated, stratum corneum samples were dried and rehydrated and then analysed.

DNA fragmentation induced in lymphocytes by gamma irradiation or dexamethasone: inhibition by diethyldithiocarbamate (DTC), potentiated by zinc.

Apoptosis is a process of physiological cell death characterized by DNA fragmentation, chromatin condensation, loss of membrane asymmetry and cell lethality. In the present study, apoptosis induced in thymocytes by dexamethasone or gamma irradiation is evaluated by flow cytometry, gel electrophoresis and other techniques. Treatment of thymocytes with DTC or zinc shows that these products can inhibit radiation- or dexamethasone-induced apoptosis.

Myocardial kinetics of TcN-NOET: a neutral lipophilic complex tracer of regional myocardial blood flow.

Unlabelled: [Bis (N-etoxy, N ethyl dithiocarbamato) nitrido] 99mTc (V) (TcN-NOET) is a new neutral lipophilic myocardial imaging agent proposed for clinical use for detecting coronary artery disease. We studied the relation between myocardial retention of TcN-NOET and myocardial blood flow (MBF) in a canine model.

Methods: A wide range of MBF was induced by partial regional coronary occlusion and dipyridamole infusion (protocols 1,2 and 3).

Ventilatory and locomotory activities in anoxia and subsequent recovery of epigean and hypogean crustaceans.

Locomotory and ventilatory responses to severe hypoxia and subsequent recovery were investigated in 3 amphipod crustaceans: 2 hypogean species (1 interstitial species Niphargus rhenorhodanensis and 1 karstic species N. virei) and 1 epigean species (Gammarus fossarum), and in an epigean population of 1 isopod crustacean (Asellus aquaticus). CNS distribution and overexpression of neurofilament light proteins (NF-L) in mice transgenic for the human NF-L: aberrant accumulation in thalamic perikarya.

Light microscopic immunocytochemistry with monoclonal antibodies recognizing both murine and human light neurofilament proteins (mNF-L and hNF-L) or hNF-L only was used to examine the distribution of NF-L in the CNS of adult mice, normal or transgenic for the human gene. In normal mice, major fiber bundles were immunoreactive to the first antibody, with few exceptions such as the internal capsule, anterior commissure, and corpus callosum. Strong immunoreactivity was also present in the perikarya of motoneurons in the spinal cord and brainstem, as well as in other brainstem nuclei.

Popliteal artery entrapment syndrome. Anatomic and embryologic bases, diagnostic and therapeutic considerations following a series of 15 cases with a review of the literature.

We report 15 examples of popliteal artery entrapment syndrome observed in 11 patients. The anatomical causes were as follows: in one case, the popliteal artery presented an aberrant course medially to the medial head of the gastrocnemius muscle. In 5 cases, there was a small fibrous band linking the medial head of the gastrocnemius muscle to the lateral condyle and crossing behind the popliteal artery; in 5 cases this anomaly was also found in association with an abnormally high and/or internal insertion of the medial head of gastrocnemius muscle.

Recognition and elimination of senescent erythrocytes: implication of antibodies specific for malonic dialdehyde-protein adducts, as demonstrated by flow cytometry.

Many different hypotheses have been formulated about the mechanisms of specific recognition of senescent red blood cells (RBC). It is usually assumed that novel epitopes appear on RBC membranes during ageing and are responsible for recognition of aged RBC by antibodies, which is followed by binding to mononuclear phagocytes and then phagocytosis. But these age-related epitopes have not so far been identified.

Typhoid fever in New York City, 1980 through 1990.

Background: Although typhoid fever incidence decreased in the 1960s and 1970s in New York City and elsewhere, it did not disappear. In this article, trends associated with various modes of transmission of Salmonella typhi in New York City patients are described.

Methods: Typhoid fever surveillance reports from 1980 to 1990 were reviewed for clinical, demographic, and epidemiologic characteristics.

Immunization of mice with proteins reacted with malonic dialdehyde (MDA): comparison between autologous and heterologous modified proteins.

In previous experiments, rabbits were injected with heterologous proteins reacted with MDA, and produced antibodies cross-reacting with other MDA-modified proteins (MPr), but not with the corresponding native ones (Pr). It was concluded that these antibodies (AbAIP) recognized epitopes including 1-amino-3-imino-propene (AIP) bridges resulting from reactions of MDA with primary amino groups of proteins. In the present work, mice were injected with autologous MDA-modified albumin (MAI) or with heterologous MPr.

Cross-level analysis of the influence of local union structure on women's and men's union commitment.

The authors tested the influence of structural features of local unions (N = 43) on members' union commitment (N = 539) as moderated by members' gender. By using a cross-level analysis, they identified local structure with officers' ratings of innovation, formalization, and centralization. An anticipated interaction between gender and innovation was not found.

Linkage disequilibrium analysis of childhood-onset spinal muscular atrophy (SMA) in the French-Canadian population.

Spinal muscular atrophy (SMA) is, after Duchenne muscular dystrophy, the most common neuromuscular disorder in childhood. The gene responsible for childhood SMA has been mapped to the q11.2-q13.

Regional uptake of [123I]-16-iodo3-R,S-methyl hexadecanoic acid in patients with myocardial infarction. Comparison with thallium 201 uptake and wall motion.

Thirty patients with recent myocardial infarction were intravenously injected with the modified fatty acid [123I]16-iodo-3R,S-methyl hexadecanoic acid (MIHA) at peak exercise to quantify viable myocardium after infarction. The results were analysed visually and quantitatively and compared with those obtained after injection of thallium 201 at peak exercise with single-photon emission computed tomography (SPECT) imaging immediately and 4 hours later. Ventriculography was used to study regional wall motion in all patients.

Restoration of postburn impaired lymphocyte responsiveness by nonsteroidal anti-inflammatory drugs is independent of prostaglandin E2 inhibition.

Prostaglandin E2 (PGE2) has been implicated in postburn immunosuppression, which is responsible for septic complications. In the present work, seven non-steroidal anti-inflammatory drugs (NSAIDs), differing by their capacity to inhibit the cyclooxygenase pathway, were compared for their ability to restore T lymphocyte proliferative responses evaluated 4 days after thermal injury in rats. Salicylic acid, 5-aminosalicylic acid, and niflumic acid, given daily, fully restored spleen cell responses to concanavalin A (Con A) and phytohemagglutinin.

Dacron patch/tube graft: a preliminary study.

A Dacron patch/tube graft was developed specifically to avoid technical complications associated with lateral aortic anastomoses of medium-diameter grafts. This new prosthetic device has now been used in 15 patients without complications. It seems to be particularly useful in patients with thick-walled aortas.

Genetic epidemiology of sensorimotor polyneuropathy with or without agenesis of the corpus callosum in northeastern Quebec.

Sensorimotor polyneuropathy with or without agenesis of the corpus callosum (McKusick number 218000) is a disorder that has a high frequency in Saguenay-Lac-St-Jean (SLSJ), a geographically isolated region of north-eastern Quebec. The incidence at birth and the carrier rate were estimated, respectively, at 1/2117 liveborns and 1/23 inhabitants. Remote consanguinity was found in several polyneuropathic families while the mean kinship coefficient was 2.

With the rapid increase in literacy during the second half of the 19th century, impersonal and variable sources of information were added to the oral discourse on health and cleanliness for growing segments of the population. Based on a Swiss sample of seventy popular journals and calendars, this article analyses the historical context, the means and effects of this process. It argues that the texts on medical hygiene in popular literature confirmed the social values of bourgeois classes and, at the same time, formed a cultural force of their own.

Immunization of rabbits with proteins reacted with malonic dialdehyde (MDA): kinetics and specificity of the immune response.

MDA-modified casein, lysozyme or polylysine (MC, ML and MP respectively), was intradermically injected to rabbits in the presence of complete Freund's adjuvant (cFA). Two other animal sets received either cFA alone, or MDA alone. MDA, cFA and MP did not induce any antibody response.
