Article Synopsis
  • Pharmacogenomic markers in HLA coding genes are linked to drug hypersensitivity reactions, particularly with the drug allopurinol.
  • A review found a strong association between the HLA-B*5801 allele and severe cutaneous adverse drug reactions (cADRs) across multiple ethnic groups, although the link for less severe reactions was inconsistent.
  • Future research should investigate more common, milder cADRs from allopurinol and explore the potential impact of rare genetic variants as predictors of these conditions.

Background: Aortic medial calcification is a cellular-regulated process leading to arterial stiffness. Although epidemiological studies have suggested an association between the saturation of fatty acids (FA) and arterial stiffness, there is no evidence that saturated FA can induce arterial calcification. This study investigated the capacity of palmitic acid (PA) to induce medial calcification and the signaling pathway(s) implicated in this process.
