Since designer cells are attracting much attention as a new modality in gene and cell therapy, it would be advantageous to develop synthetic receptors that recognize artificial ligands and activate solely signaling molecules of interest. In this study, we refined the construction of our previously developed minimal engineered receptors (MERs) to avoid off-target activation of STAT5 while maintaining on-target activation of signaling molecules corresponding to tyrosine motifs. Among the myristoylated, cytoplasmic, and transmembrane types of MERs, the cytoplasmic type had the highest signaling efficiency, although there was off-target activation of STAT5 upon ligand stimulation.

The use of a "direct PCR" DNA polymerase enables PCR amplification without any prior DNA purification from blood samples due to the enzyme's resistance to inhibitors present in blood components. View Article and Find Full Text PDFFluoroalkyl end-capped vinyltrimethoxysilane oligomer [R(F)-(VM)(n)-R(F)] underwent the sol-gel reaction under alkaline conditions in the presence of anatase titanium oxide nanoparticles (an-TiO(2)) in tetrahydrofuran to give the corresponding fluorinated oligomer/anatase titanium oxide nanocomposites [R(F)-(VM-SiO(2))(n)-R(F)/an-TiO(2)]. Crystalline structure of an-TiO(2) in the nanocomposites thus obtained was found to keep completely its structure without phase transformation to rutile even after calcination at 1000°C, although crystalline structure of the original an-TiO(2) nanoparticles underwent a complete phase transformation to the rutile under similar conditions. Interestingly, R(F)-(VM-SiO(2))(n)-R(F)/an-TiO(2) nanocomposites before and after calcination at 1000°C exhibited the similar photocatalytic activity for the decolorization of methylene blue under UV light irradiation.

The nanostructures of the layer were characterized by transmission electron microscopy and found to be comparatively dense and predominantly composed of c-axis oriented grains. Methods: Sixty patients scheduled to receive combined spinal-epidural anesthesia were randomly allocated to receive either spinal anesthesia first (group CSEA) or epidural lidocaine first (group Epi). Each group was divided into three subgroups in which the site of epidural cannulation and spinal tap were separated by one, three, or five interspaces (sets I, II, and III, respectively).
We identified seven distinct subtypes of enterohemorrhagic Escherichia coli (EHEC) O157:H7 isolates that were derived from sporadic cases and outbreaks from multiple prefectures in Japan in 2005. A surveillance system utilizing pulsed-field gel electrophoresis (PFGE), PulseNet Japan, was used. Some strains showed indistinguishable PFGE patterns using another restriction enzyme (BlnI or SpeI) in each subtype of EHEC O157:H7 isolates that were routinely subtyped by the XbaI PFGE pattern.
