The role of sphingosine 1-phosphate (S1P) and its sphingosine-1-phosphate receptors (S1PRs) in non-alcoholic steatohepatitis (NASH) is unclear. We aimed to analyze the role of S1P/S1PRs in a Melanocortin-4 receptor (Mc4r)-deficient NASH murine model using FTY720, the functional antagonist of S1PR1, S1PR3, S1PR4, and S1PR5, and JTE-013, the antagonist of S1PR2. We observed that, compared to that in the control, the mRNA of S1pr1 tended to decrease, whereas those of S1pr2 and S1pr3 significantly increased in Mc4r-knockout (KO) mice subjected to a Western diet (WD).

Reflex responses play an important role in gravity resistance, balance maintenance and the reaction to perturbation during human walking and standing upright. In this study, aiming at the realization of artificial reflex for paralyzed people, whose reflexive system was also impaired to a certain degree because of the weakened afferent neural pathway, the reflex responses of normal subjects were measured and analyzed. Charcot joint is a disease that often occurs in patients with diabetes mellitus, tabes dorsalis, syringomyelia, chronic alcoholism, leprosy, trauma, or infection after fractures and dislocations. The treatment for Charcot joint has various complications, such as skin lesions, infections, and delayed union. We present our experience with a male patient who developed Charcot joint-like changes without diabetes mellitus or any other disease after an ankle fracture due to minor trauma.

In vitro dynamic simulation of knee flexion was performed to quantify knee kinematics for a mobile bearing prosthesis that allows the tibial insert to translate and rotate with respect to the baseplate. Six cadaver knees were tested in the intact state, after implanting a fixed platform prosthesis, and after implanting a mobile bearing prosthesis. The mobile bearing prosthesis significantly increased the tibial internal rotation and medial shift compared with the intact knee, near 90 degrees of flexion.

Isolated posterior cruciate ligament injuries usually are treated nonoperatively, although some patients remain symptomatic, and degenerative changes within the patellofemoral joint and the medial compartment of the tibiofemoral joint have been seen in followup studies. In vitro simulation of knee squatting was done to quantify the influence of the posterior cruciate ligament on tibiofemoral and patellofemoral kinematics. For five knee specimens, knee kinematics were measured before and after sectioning the posterior cruciate ligament, and compared using a Wilcoxon signed rank test.
