Article Synopsis
  • - Thrombotic thrombocytopenic purpura (TTP) is a serious disease linked to low ADAMTS13 activity, which is crucial for blood clot regulation; it can be congenital (due to genetic issues) or acquired (due to autoantibodies).
  • - Conventional treatments like plasma exchange can be risky, but a new drug called caplacizumab helps prevent clots and may improve survival rates, even allowing for treatment without plasma exchange.
  • - Current strategies for congenital TTP involve using fresh-frozen plasma, but ongoing developments in therapies like recombinant ADAMTS13 and gene therapy could provide better long-term solutions.

Pure red cell aplasia (PRCA) associated with erythropoiesis-stimulating agents (ESAs), which were first reported in 1998, usually occurs with subcutaneous administration of epoetin alfa (Eprex). Improvements in ESA storage, handling, and administration methods have reduced the PRCA incidence. Continuous erythropoietin receptor activator (CERA) is a third-generation ESA that is rarely reported to induce PRCA.
