Moyamoya is a rare chronic brain vascular disease with a set of potential life-threatening consequences due to a high probability of stroke occurrence. Here we present a case of a patient with moyamoya disease, who has been dynamically observed over more than 4 years after the first manifestation of her symptoms. In this study we report fMRI and DTI findings obtained at the final follow-up point, four years after two revascularization surgeries were successfully performed.

Article Synopsis
  • Ribosome biogenesis starts with RNA polymerase I (Pol I), which has been linked to longevity, but its broader effects were not fully understood before this study.* -
  • The research indicates that reducing Pol I activity in Caenorhabditis elegans enhances lipid metabolism and mitochondrial function, promoting increased lifespan and better energy management, which is also seen in human cells.* -
  • Interestingly, boosting pre-rRNA synthesis improves growth in young nematodes but leads to faster aging, highlighting that managing Pol I activity can significantly affect lifespan and healthspan, even when the intervention is applied later in life.*

Parkinson's Disease (PD) is the second most common neurodegenerative disorder. The pathological hallmark of PD is loss of dopaminergic neurons and the presence of aggregated α-synuclein, primarily in the substantia nigra pars compacta (SNpc) of the midbrain. However, the molecular mechanisms that underlie the pathology in different cell types is not currently understood.

Background: Primary trigeminal neuralgia (PTN) is a prevalent chronic pain disorder whose pathogenesis is not limited to the trigeminal system. Despite the significant advances in uncovering underlying mechanisms, there is a paucity of comprehensive and consistent data regarding the role of white matter throughout the entire brain in PTN.

Methods: We performed a prospective case-control study.

Introduction: Surgical revascularization is very effective in patients with moyamoya angiopathy (MMA) and leads to improvements in cortical perfusion parameters. However, changes in white matter hemodynamics are still underestimated. To date, only a few studies have examined brain perfusion changes within deep white matter after bypass surgery in patients with MMA.

Cognitive decline is a common pathological outcome during aging, with an ill-defined molecular and cellular basis. In recent years, the concept of inflammaging, defined as a low-grade inflammation increasing with age, has emerged. Infiltrating T cells accumulate in the brain with age and may contribute to the amplification of inflammatory cascades and disruptions to the neurogenic niche observed with age.

Antiphospholipid syndrome (APS) is an autoimmune thrombophilia that is characterised by thrombosis and obstetric complications in the presence of antiphospholipid antibodies (aPL). Pregnancy complications remain a challenging problem for patients with APS, especially during the first trimester. Although natural killer (NK) cells constitute up to 70% of decidual lymphocytes during the first trimester, their contribution to early pregnancy loss in APS is largely unknown.

Article Synopsis
  • Microgravity conditions, utilized since the early 1980s, enhance protein crystallization by minimizing convection and creating a sedimentation-free environment, leading to higher quality crystals.
  • Experiments aboard the International Space Station (ISS) have shown that although crystal growth rates are slower in microgravity, this environment allows for extended, detailed studies on macromolecular transport and impurity incorporation in growing crystals.
  • Findings reveal that crystals grown in microgravity incorporate fewer impurities and often display better quality, providing insights for predicting which proteins might benefit most from this crystallization method.

Purpose: To evaluate the mechanisms of how therapeutic upregulation of the transcription factor, CCAAT/enhancer-binding protein alpha (C/EBPα), prevents tumor progression in patients with advanced hepatocellular carcinoma (HCC) and in different mouse tumor models.

Experimental Design: We conducted a phase I trial in 36 patients with HCC (NCT02716012) who received sorafenib as part of their standard care, and were given therapeutic C/EBPα small activating RNA (saRNA; MTL-CEBPA) as either neoadjuvant or adjuvant treatment. In the preclinical setting, the effects of MTL-CEBPA were assessed in several mouse models, including BNL-1ME liver cancer, Lewis lung carcinoma (LLC), and colon adenocarcinoma (MC38).

Familial Mediterranean fever (FMF) is caused by pyrin-encoding gene mutations and characterized by the self-limiting periods of intense inflammation, which are mainly mediated by a massive influx of polymorphonuclear neutrophils (PMNs) into the inflamed sites. Perturbation of actin polymerization by different pathogens was shown to activate the pyrin inflammasome. Our aim was to test whether cytoskeletal dynamics in the absence of pathogens may cause abnormal activation of PMNs from FMF patients.

We demonstrate a novel concept, to the best of our knowledge, for reproduction of the target movement without the frame-by-frame display technique elaborated by the Lumiere brothers. The specially designed elements-apodizing filters with axially quadratic transmittance of radiation, are used to continually record a target position change in time. The recording system deals with nonmatrix detectors and requires only one-step conversion of light energy into an electrical signal to monitor the target under study.

Antiphospholipid syndrome (APS) is the most common cause of acquired thrombophilia and recurrent spontaneous miscarriages associated with extended persistence of antiphospholipid antibodies (aPL). How circulating aPL and high-17β-estradiol (E2) environment contribute to the pregnancy complications in APS is poorly defined. Therefore, we aimed to analyse whether E2 could be responsible for the immune cell hyperactivation in aPL- positive (lupus anticoagulant, anti-cardiolipin, anti-β2-glycoprotein) in women.

In this paper, we describe a novel concept, to the best of our knowledge, to monitor changes in the landscape with two arbitrarily distributed substances. To record signals from the mock-up scene, nonmatrix detectors are used, which work in the linear sensitivity range. The formulas that describe the parameters of distributed substances are derived.

Although the cerebral cortex is organized into six excitatory neuronal layers, it is unclear whether glial cells show distinct layering. In the present study, we developed a high-content pipeline, the large-area spatial transcriptomic (LaST) map, which can quantify single-cell gene expression in situ. Screening 46 candidate genes for astrocyte diversity across the mouse cortex, we identified superficial, mid and deep astrocyte identities in gradient layer patterns that were distinct from those of neurons.
