High dose-intensive or infusional intermediate-dose immunochemotherapy is highly effective treatment for Burkitt lymphoma irrespective of human immunodeficiency virus (HIV) infection. However, toxicities of these regimens are relevant, especially in older adults and elderly patients. The prospective multicenter BURKIMAB14 trial included four to six blocks of immunochemotherapy according to stage (localized: 1 and 2 non-bulky; advanced: 2 bulky, 3, 4) and age, with dose reduction in patients >55 years old.

Introduction: The prognosis of relapsed B cell precursor acute lymphoblastic leukemia (B-ALL) is poor and few patients can be successfully rescued with conventional therapies. Inotuzumab ozogamicin (IO), an antibody against the CD22 antigen linked to calicheamicin, has been approved as a rescue treatment in relapsed/refractory (R/R) B-ALL.

Patients And Methods: This was an observational, retrospective, multicenter study of adult patients included in the Spanish program of compassionate use of IO in centers from the PETHEMA group (Programa Español de Tratamientos en Hematología).

Article Synopsis
  • The study focuses on a new DNA-based next-generation sequencing (NGS) method for detecting and monitoring BCR::ABL1 KD mutations in patients with chronic myeloid leukemia (CML) and Philadelphia-positive acute lymphoblastic leukemia (ALL).
  • This NGS method shows high sensitivity (1.0E-4) and a strong correlation with an established RNA-based method, indicating that it can effectively quantify ABL1 mutations.
  • The study analyzed 162 samples from 129 patients, identifying significant mutations in both CML and B-ALL cases, including instances where mutations were detected at very low disease levels.

Article Synopsis
  • The study investigates kidney dysfunction patterns in patients with acute decompensation of cirrhosis, both with and without acute-on-chronic liver failure (ACLF), involving 639 patient admissions over a 3-month follow-up period.
  • A significant 92% of ACLF patients experienced acute kidney injury (AKI), contrasting with only 35% of non-ACLF patients, highlighting the severe impact of ACLF on kidney health.
  • Outcomes for patients with AKI in the ACLF group were notably poor, with only 54% recovering kidney function and a 3-month survival rate of 51%, compared to much better outcomes in non-ACLF patients; factors such as

Background And Aims: Stigmatization is a well-documented problem of some diseases. Perceived stigma is common in alcohol-related liver disease and hepatitis C, but little information exists on stigma in patients with non-alcoholic fatty liver disease (NAFLD). Aim of the study was to investigate frequency and characteristics of perceived stigma among patients with NAFLD.

Whether tubular injury develops in patients with acute kidney injury owing to hepatorenal syndrome (AKI-HRS) is controversial. We performed repeated measurements of biomarkers of tubular injury during a 14-day period in 60 patients with cirrhosis and AKI (34 with AKI-HRS meeting the classical definition of type 1 HRS and 26 with AKI owing to acute tubular necrosis, AKI-ATN). Nineteen of 34 patients had resolution of AKI-HRS, while the remainder had persistent AKI-HRS.

The prognosis of t(1;19)(q23;p13)/transcription factor 3-pre-B-cell leukaemia homeobox 1 (TCF3-PBX1) in adolescent and adult patients with acute lymphoblastic leukaemia (ALL) treated with measurable residual disease (MRD)-oriented trials remains controversial. In the present study, we analysed the outcome of adolescent and adult patients with t(1;19)(q23;p13) enrolled in paediatric-inspired trials. The patients with TCF3-PBX1 showed similar MRD clearance and did not have different survival compared with other B-cell precursor ALL patients.

Article Synopsis
  • Decompensated cirrhosis (DC) is a serious liver condition that can lead to high death rates, especially when it turns into acute-on-chronic liver failure (ACLF).
  • Researchers wanted to see if measuring a protein called L-FABP in urine could help predict which DC patients are at higher risk of complications or death.
  • The study found that higher urine L-FABP levels were linked to worse outcomes in patients, making it a potential tool for doctors to assess patient risks.

The potential prognostic value of conventional karyotyping in adult T-cell acute lymphoblastic leukemia (T-ALL) remains an open question. We hypothesized that a modified cytogenetic classification, based on the number and type of cytogenetic abnormalities, would allow the identification of high-risk adult T-ALL patients. Complex karyotype defined by the presence of ≥3 cytogenetic alterations identified T-ALL patients with poor prognosis in this study.

The need for allogeneic hematopoietic stem cell transplantation (allo-HSCT) in adults with Philadelphia chromosome-negative (Ph-) acute lymphoblastic leukemia (ALL) with high-risk (HR) features and adequate measurable residual disease (MRD) clearance remains unclear. The aim of the ALL-HR-11 trial was to evaluate the outcomes of HR Ph- adult ALL patients following chemotherapy or allo-HSCT administered based on end-induction and consolidation MRD levels. Patients aged 15 to 60 years with HR-ALL in complete response (CR) and MRD levels (centrally assessed by 8-color flow cytometry) <0.

BCR-ABL1-like B-cell precursor acute lymphoblastic leukemia (BCP-ALL) remains poorly characterized in adults. We sought to establish the frequency and outcome of adolescent and adult BCR-ABL1-like ALL using a novel RNA-Seq signature in a series of patients with BCP-ALL. To this end, we developed and tested an RNA-Seq custom panel of 42 genes related to a BCR-ABL1-like signature in a cohort of 100 patients with BCP-ALL and treated with risk-adapted ALL trials.
