Anemia of Inflammation is a prevalent co-morbidity in patients with chronic inflammatory disorders. Inflammation causes hypoferremia and iron-restricted erythropoiesis by limiting Ferroportin (FPN)-mediated iron export from macrophages that recycle senescent erythrocytes. Macrophage cell surface expression of FPN is reduced by hepcidin-induced degradation and/or by repression of FPN (Slc40a1) transcription via cytokine and Toll-like receptor (TLR) stimulation.

Article Synopsis
  • About one-third of Portuguese patients with congenital hypogonadotropic hypogonadism (CHH) have a genetic cause linked to 10 different genes, with a small percentage showing oligogenic inheritance.
  • CHH leads to delayed puberty and infertility due to issues with hormones like GnRH, LH, and FSH.
  • In a study involving 81 patients and 263 controls, genetic screening helped identify pathogenic variants, but many variants classified as uncertain complicate the understanding of the disease causes.

Skin lesion datasets used in the research are highly imbalanced; Generative Adversarial Networks can generate synthetic skin lesion images to solve the class imbalance problem, but it can result in bias and domain shift. Domain shifts in skin lesion datasets can also occur if different instruments or imaging resolutions are used to capture skin lesion images. The deep learning models may not perform well in the presence of bias and domain shift in skin lesion datasets.

Article Synopsis
  • The study aimed to investigate whether smoking impacts the severity of acute radiation toxicity in head and neck cancer patients undergoing radiotherapy.
  • Out of 102 patients studied, severe acute toxicities like mucositis and dysphagia occurred in various percentages, but no significant link was found between smoking status and these toxicities during treatment.
  • Factors like concurrent chemotherapy and different radiation fractionation schedules were identified as significant contributors to the occurrence of severe acute toxicity, rather than smoking.

ALK-positive NSCLC patients demonstrate initial responses to ALK tyrosine kinase inhibitor (TKI) treatments, but eventually develop resistance, causing rapid tumor relapse and poor survival rates. Growing evidence suggests that the combination of drug and immune therapies greatly improves patient survival; however, due to the low immunogenicity of the tumors, ALK-positive patients do not respond to currently available immunotherapies. Tumor-associated macrophages (TAMs) play a crucial role in facilitating lung cancer growth by suppressing tumoricidal immune activation and absorbing chemotherapeutics.

The liver plays a crucial role in maintaining systemic iron homeostasis through iron storage, sensing of systemic iron needs, and production of the iron-regulatory hormone hepcidin. While mice are commonly used as models for studying human iron homeostasis, their liver structure differs significantly from humans. Since the mouse liver is structured in six separated lobes, often, the analysis of a single defined lobe is preferred due to concerns over data reproducibility between experimental cohorts.

Purpose: The endogenous hypercortisolism that characterizes Cushing's syndrome (CS) is associated with a state of hypercoagulability that significantly increases the risk of thromboembolic disease, especially, venous events. Despite this certainty, there is no consensus on the best thromboprophylaxis strategy (TPS) for these patients. Our aim was to summarize the published data about different thromboprophylaxis strategies, and to review available clinical tools assisting thromboprophylaxis decision making.

Skin cancer is one of the primary causes of death globally, and experts diagnose it by visual inspection, which can be inaccurate. The need for developing a computer-aided method to aid dermatologists in diagnosing skin cancer is highlighted by the fact that early identification can lower the number of deaths caused by skin malignancies. Among computer-aided techniques, deep learning is the most popular for identifying cancer from skin lesion images.

Article Synopsis
  • About 15% of patients with common variable immunodeficiency (CVID) develop a small intestinal enteropathy that resembles celiac disease but originates from a different cause, sometimes linked to chronic norovirus infection.
  • Analysis of duodenal biopsies from CVID patients showed that the development of villous atrophy (VA) is associated with a shortage of IgA plasma cells and an imbalance in T helper cells, along with inflammation driven by different types of interferons.
  • The study indicates that interferon signaling and T-cell activity increase in severity from mild to severe stages of CVID enteropathy, with norovirus infection intensifying inflammatory responses, paving the way for better-targeted treatment options.

The majority of pituitary adenomas occur in a sporadic context, and in the absence of known genetic predisposition. Three common variants at the (rs2359536), (rs10763170) and (rs17083838) loci were previously associated with sporadic pituitary adenomas in the Han Chinese population, but these findings have not yet been replicated in any other population. The aim of this case-control study was to assess if these variants are associated with susceptibility to sporadic pituitary adenomas in the Portuguese population.
