Article Synopsis
  • Daphnetoxin is a diterpene found in Thymelaeaceae plants and is more toxic to mitochondria than daphnoretin, which is a bis-coumarin from multiple plant families.
  • Both compounds show potential anti-cancer properties, but exhibit toxicity to Wistar rat liver mitochondria and all tested cell lines at concentrations over 100 microM.
  • The study found that while both compounds inhibited cell proliferation, the mechanisms may be partially mitochondrial-independent, suggesting a need for further evaluation of their safety and pharmacological benefits.

The effects of daphnetoxin on isolated rat liver mitochondria and freshly isolated rat hepatocytes were investigated. Daphnetoxin (in the microM range) increased mitochondrial state 4 respiration and decreased both state 3 and FCCP-uncoupled respiration. The transmembrane potential was strongly depressed by daphnetoxin in a concentration-dependent manner.

A new diterpene tetraester, from the jatrophane family, and two new diterpene triesters, with a lathyrane skeleton, have been isolated from the chloroform extract of the roots of Euphorbia hyberna L. The structures of these compounds have been established by spectroscopic methods, including 2D NMR experiments.
