A dramatic improvement is reported in the stability of colloidal particles when stabilizing surface grafts are systematically shortened from small polymers to single monomers. The colloidal dispersions consist of fluorinated latex particles, exhibiting a weak van der Waals attraction, with grafted steric layers of poly(ethylene glycol) (PEG) of different chain lengths. Using an effective salting-out electrolyte, NaCO, particle aggregates are detected above a threshold salt concentration that is independent of the particle concentration.

Fluorinated spheres with grafted poly(ethylene glycol) (PEG) have been synthesized using a semi-batch emulsion polymerization in which the initiator is fed slowly to the reaction. In this way, PEG-grafted colloidal spheres can be fabricated with varying PEG chain length, different cores and varying degrees of crosslinking. The resulting batches have been characterized using disc centrifuge photosedimentometry and small-angle X-ray scattering.

The phase behavior of lactoferrin has been studied as a function of concentration at a pH and ionic strength where lactoferrin is known to interact effectively via a patch-patch attraction. In contrast to isotropic attractive potentials, the directional attraction gives rise to a different phase or solution behavior. At low concentrations, the protein dimerizes.

Self-association of the protein lactoferrin is studied in solution using small-angle X-ray scattering techniques. Effective static structure factors have been shown to exhibit either a monotonic or a nonmonotonic dependence on protein concentration in the small wavevector limit, depending on salt concentration. The behavior correlates with a nonmonotonic dependence of the second virial coefficient on salt concentration, such that a maximum appears in the structure factor at a low protein concentration when the second virial coefficient is negative and close to a minimum.

Aqueous dispersions of fluorinated colloidal spheres bearing grafted poly(ethylene glycol) (PEG) are studied as a function of salt and particle concentration with the aim of improving the understanding of interactions among polymer-grafted particles. These dispersions can sustain large concentrations of salt, but crystals nucleate in dilute dispersions when a sufficient Na2CO3 concentration is reached, which is attributed to the presence of attractions between particles. On further increasing the Na2CO3 concentration, the solvent is rapidly cleared of particles.

A laboratory-scale small-angle X-ray scattering instrument with pinhole collimation has been used to assess smearing effects due to instrumental resolution. A new, numerically efficient method to smear ideal model intensities is developed and presented. It allows for directly using measured profiles of isotropic but otherwise arbitrary beams in smearing calculations.

The intronic SNP rs7903146 in the T-cell factor 7-like 2 gene (TCF7L2) is the common genetic variant most highly associated with Type 2 diabetes known to date. The risk T-allele is located in an open chromatin region specific to human pancreatic islets of Langerhans, thereby accessible for binding of regulatory proteins. The risk T-allele locus exhibits stronger enhancer activity compared to the non-risk C-allele.

Static light scattering (SLS) combined with structure-based Monte Carlo (MC) simulations provide new insights into mechanisms behind anisotropic, attractive protein interactions. A nonmonotonic behavior of the osmotic second virial coefficient as a function of ionic strength is here shown to originate from a few charged amino acids forming an electrostatic attractive patch, highly directional and complementary. Together with Coulombic repulsion, this attractive patch results in two counteracting electrostatic contributions to the interaction free energy which, by operating over different length scales, is manifested in a subtle, salt-induced minimum in the second virial coefficient as observed in both experiment and simulations.

Fluorinated core-shell spheres have been synthesized using a novel semibatch emulsion polymerization protocol employing slow feeding of the initiator. The synthesis results in aqueous dispersions of highly monodisperse spheres bearing a well-defined poly(ethylene glycol) graft (PEGylation). Measurements are consistent with the synthesis achieving a high grafting density that moreover consists of a single PEG layer with the polymer significantly elongated beyond its radius of gyration in bulk.

We have investigated the temperature induced micelle-to-vesicle transition in a binary non-ionic surfactant/water system, for which the spontaneous curvature decreases with increasing temperature. Temperature jumps with variable rate were performed in a microwave oven, from a micellar phase at 5°C to a lamellar phase region at 35°C, passing a liquid-liquid two phase region where dilute and concentrated micellar solutions coexist. It is shown that the obtained vesicle size decreases with increasing heating rate through this two phase region.

We present an experimental study of vesicle fusion using light and neutron scattering to monitor fusion events. Vesicles are reproducibly formed with an extrusion procedure using an single amphiphile triethylene glycol mono-n-decyl ether in water. They show long-term stability for temperatures around 20 degrees C, but at temperatures above 26 degrees C we observe an increase in the scattered intensity due to fusion.
