Molecules that induce novel interactions between proteins hold great promise for the study of biological systems and the development of therapeutics, but their discovery has been limited by the complexities of rationally designing interactions between three components, and because known binders to each protein are typically required to inform initial designs. Here, we report a general and rapid method for discovering α-helically constrained (Helicon) polypeptides that cooperatively induce the interaction between two target proteins without relying on previously known binders or an intrinsic affinity between the proteins. We show that Helicons are capable of binding every major class of E3 ubiquitin ligases, which are of great biological and therapeutic interest but remain largely intractable to targeting by small molecules.

The objectives of this study were to investigate whether blastocoel collapse before vitrification induced by laser improves the cryo-survivability of buffalo in-vitro-fertilized (IVF) blastocysts and whether laser assisted hatching (LAH) promotes hatchability of fresh and frozen-thawed IVF blastocysts. The expanded blastocysts were harvested on Days 6-9 and randomly allocated into five groups as follows: (1) blastocysts were vitrified and thawed without any treatment; (2) blastocysts were vitrified after 15-20 μm zona pellucida (ZP) thinning opposite to the inner cell mass, and blastocoels were also blotted in order to outflow the blastocoelic fluid before vitrification; (3) ZP thinning was made immediately after thawing; (4) fresh blastocysts underwent LAH; and (5) as a control, fresh blastocysts without treatment. Composites of multi-walled carbon nanotubes (MWCNTs) and superparamagnetic magnetite nanoparticles, FeO@MWCNT, were synthesized in DMF in a vortex fluidic device (VFD). This involved generation of the iron oxide nanoparticles by laser ablation of bulk iron metal at 1064 nm using a pulsed laser, over the dynamic thin film in the microfluidic platform. To address the limitations of piezoelectric polymers which have a low dielectric constant andto improve their dielectric and ferroelectric efficiency for energy storage applications, we designed and characterized a new hybrid composite that contains polyvinylidene fluoride as a dielectric polymer matrix combined with graphene platelets as a conductive and barium titanite as ceramic ferroelectric fillers. Different graphene/barium titanate/polyvinylidene fluoride nanocomposite films were synthesized by changing the concentration of graphene and barium titanate to explore the impact of each component and their potential synergetic effect on dielectric and ferroelectric properties of the composite. Results showed that with an increase in the barium titanate fraction, dielectric efficiency ofthe nanocomposite was improved.
