Article Synopsis
  • A new method using elastic lipid nanovesicles (ELNs) loaded with Lantana camara extract is developed to improve the permeation of active compounds through the skin.
  • The study produced 15 different formulations and examined factors like vesicle size, surface characteristics, and the impact on skin permeation using a model resembling human skin (Start-M).
  • Results showed that smaller vesicle sizes (100-200 nm) significantly enhanced the permeation of the extract, and that the optimal surfactant ratio led to over 75% of the extract permeating within 8 hours; the ELN formulation also demonstrated a strong reduction in skin cancer cell viability.
  • Future research should focus on the long-term safety and clinical applications of ELNs

Aim: To explore parental perceptions of the consenting process and understanding of the study in a pilot randomised controlled trial wherein extremely premature infants (<29 weeks' gestation) were recruited either antenatally or by 4 h of life.

Methods: We prospectively surveyed parents who had consented, declined consent or were eligible infants in the Positioning Preterm Infants for Neuroprotection study, a low-risk intervention study in the first 72 h of life. Structured interview questions explored the process and acceptability of the consenting approach by the parents and their knowledge of the study.

  • This phase 1b study assessed the safety and effectiveness of cilofexor, a drug targeting farnesoid X receptors, in patients with compensated cirrhosis caused by primary sclerosing cholangitis over 12 weeks.
  • Patients received increasing doses of cilofexor, and while most reported side effects like pruritus and fatigue, there were no serious adverse events.
  • Results showed significant improvements in liver function markers, indicating that cilofexor may help manage cholestasis in these patients.

Konjac glucomannan (KG) is a dietary fiber hydrocolloid derived from Amorphophallus konjac tubers and is widely utilized as a food additive and dietary supplement. As a health-conscious choice, purified KG, along with konjac flour and KG-infused diets, have gained widespread acceptance in Asian and European markets. An overview of the chemical composition and structure of KG is given in this review, along with thorough explanations of the processes used in its extraction, production, and purification.

Lipid nanoparticles (LNPs) have emerged as the dominant platform for RNA delivery, based on their success in the COVID-19 vaccines and late-stage clinical studies in other indications. However, we and others have shown that LNPs induce severe inflammation, and massively aggravate pre-existing inflammation. Here, using structure-function screening of lipids and analyses of signaling pathways, we elucidate the mechanisms of LNP-associated inflammation and demonstrate solutions.
